民猪肝脏对冷暴露的生理反应和代谢组学应答

摘要 [目的]本文旨在解析猪的肝脏组织对低温环境的反应机理。[方法]利用液相色谱质谱联用(LC-MS)技术研究了慢性冷暴露和急性冷暴露对民猪肝脏组织的代谢影响。试验选用6月龄体重相近的雌性民猪9头,随机分成3组,1组为对照组,1组为慢性冷暴露组(58 d),1组为急性冷暴露组(3 d)。[结果]低温处理后,共鉴定出434个正离子和344个负离子代谢物,功能涵盖了5大类生物通路,主要聚集在新陈代谢和生物体系统2大生物过程。慢性冷暴露组共有59个差异显著的代谢物,其中32个显著上调,27个显著下调,急性冷暴露组共有17个差异显著的代谢物,其中9个显著上调,8个显著下调。组间仅有3个共同代谢物,分别为葡糖石胆酸、5-甲基尿苷和2-苯基丙酸。慢性冷暴露组的差异代谢物显著富集在37个代谢通路上,以β-丙氨酸代谢通路和铁死亡通路富集最显著,急性冷暴露组的差异代谢物显著富集在8个代谢通路上,以牛磺酸和亚牛磺酸代谢通路富集最显著。慢性冷暴露虽然导致肝脏多个生物学通路发生了显著变化,但每个通路上所富集的代谢物均很少。[结论]慢性冷暴露对民猪肝脏代谢的影响要大于急性冷暴露,冷暴露改变了肝脏的脂质代谢和糖代谢,慢性冷暴露最终会造成肝细胞出现铁死亡现象。但从代谢物变化的倍数以及所改变的通路推测,冷暴露没有对民猪的肝脏造成损伤。 [Objective]Current study was conducted to analyze the reaction mechanism of pig liver reacting to low temperature environment.[Methods]The effects of chronic or acute cold exposure on liver metabolism of Min pigs were studied by liquid chromatography-mass spectrometry(LC-MS).Nine 6-month-old female Min pigs with similar body weight were randomly divided into three groups.Group one was under chronic cold exposure for 58 d;group two was under acute cold exposure group for 3 days and group three was control group.[Results]The results showed that a total of 434 positive and 344 negative ion metabolites were identified after low temperature treatment,and their functions covered five major categories of biological pathways,mainly in the two biological processes of metabolism and biological system.In the chronic cold exposure group,59 metabolites showed significant difference,of which 32 were significantly up-regulated and 27 were down-regulated.In the acute cold exposure group,there were 17 metabolites with significant differences,of which nine were up-regulated and eight were down-regulated.There were only three common metabolites between the groups,namely glycolithocholic acid,5-methyluridine and 2-phenylpropionic acid.In the chronic cold exposure group,the differential metabolites were significantly enriched in37 metabolic pathways with the most significant enrichment in theβ-alanine metabolic pathway and Ferroptosis pathway,while in the acute cold exposure group,the differential metabolites were significantly enriched in eight pathways,in which the Taurine and hypotaurine metabolism pathways were the most enriched.Although chronic cold exposure caused significant changes in multiple biological pathways in the liver,each pathway was enriched with very few metabolites.[Conclusion]The results indicated that chronic cold exposure posed a greater effect on liver metabolism than acute cold exposure.Cold exposure changes lipid metabolism and glucose metabolism in liver,and chronic cold exposure eventually causes ferropto
