魔芋精粉酶解物对糖尿病大鼠护肝作用 被引量:3

摘要 探讨魔芋精粉酶解物的降血糖和护肝功能,为魔芋精粉酶解物的开发利用奠定基础。单次注射链脲佐菌素(streptozotocin,STZ)(45 mg/kg)建立糖尿病大鼠模型,挑选血糖值≥16.7 mmol/L的大鼠随机分为模型组、二甲双胍组[0.140 g/(kg·d)]及魔芋精粉酶解物低、中、高剂量[0.625、1.250、2.500 g/(kg·d)]组,对照组和模型组给予等体积生理盐水。连续给药6周后,测定大鼠体质量和空腹血糖水平,测定血清中谷丙转氨酶(glutamic pyruvic transaminase,ALT)、谷草转氨酶(glutamic oxaloacetic transaminase,AST)、总超氧化物歧化酶(total superoxide dismutase,T-SOD)、谷胱甘肽过氧化物酶(glutathione peroxidase,GSH-Px)活力以及丙二醛(malondialdehyde,MDA)含量,检测肝脏组织中肿瘤坏死因子(tumor necrosis factor,TNF-α)、活化B细胞κ轻链增强子的核因子(nuclear factor kappa-light-chain-enhancer of activated B cells,NF-κB)及核因子E2相关因子2(nuclear factor E2 related factor 2,Nrf)的基因表达水平并检测肝组织病理学变化。结果表明,与模型组对比,魔芋精粉酶解物组整体可以显著提高糖尿病大鼠体质量并降低血糖水平(P<0.05)、显著降低大鼠血清中ALT活性、AST活性和MDA含量(P<0.05)并显著提升大鼠血清中T-SOD活性和GSHPx活性(P<0.05),可以显著降低大鼠肝组织中TNF-α和NF-κB的基因表达量(P<0.05)。同时相比二甲双胍组,魔芋精粉酶解物组整体可以提升大鼠肝组织中Nrf的基因表达量。魔芋精粉酶解物对肝脏组织病理学损伤起到改善效果,可以改善STZ诱导的糖尿病肝损伤,机制可能与降血糖、降低脂质过氧化水平、提高机体抗氧化能力以及抗炎作用有关。 The present study explored the hypoglycemic and liver-protecting effects of konjac powder enzymatic hydrolysate to lay a foundation for its development and utilization. The diabetes model was induced by single injection of streptozotocin(STZ)at 45 mg/kg in rats. The rats with blood glucose ≥16.7 mmol/L were randomly divided into a model group,a metformin group [0.140 g/(kg·d)] and low-,medium-and high-dose konjac powder enzymatic hydrolysate groups [0.625,1.250,2.500 g/(kg·d)]. The rats in the control group and the model group were given an equal volume of normal saline. After continuous administration for six weeks,the body weight,fasting blood glucose levels,glutamic pyruvic transaminase(ALT),glutamic oxaloacetic transaminase(AST),total superoxide dismutase(T-SOD),glutathione peroxidase(GSH-Px)activity and malondialdehyde(MDA)content in the serum,gene expression of tumor necrosis factor(TNF-α),nuclear factor kappalight-chain-enhancer of activated B cells(NF-κB)and nuclear factor E2 related factor 2(Nrf)in liver tissues were detected,and histopathological changes in the liver were observed. The results showed that compared with the model group,konjac powder enzymolysis group could significantly improve the body weight and reduce the blood glucose level of diabetic rats(P<0.05). It could significantly reduce the activity of ALT(P<0.05),AST(P<0.05)and MDA content(P<0.05),and significantly increase the activity of T-SOD(P<0.05)and GSH-Px(P<0.05)in serum of rats. The expression levels of TNF-α and NF-κB in liver tissue of rats were significantly decreased(P<0.05). At the same time,compared with metformin group,konjac extract hydrolysate group could improve the expression of Nrfgene in rat liver tissue. The enzymatic hydrolysate of konjac powder can improve the histopathological injury of liver. The enzymatic hydrolysate of konjac powder can improve STZ-induced diabetic liver injury,and the mechanism may be related to the lowering of blood glucose,the reduction of lipid peroxidation level,and improvement of a
