摘要
目的建立HPLC法同时测定温经活血搽剂中葛根素、龙胆苦苷、阿魏酸、蛇床子素、异欧前胡素5种成分的含量。方法采用Eclipse XDB-C_(18)色谱柱(4.6 mm×250 mm,5μm);流动相:乙睛-0.1%磷酸,梯度洗脱;体积流量:1.0 ml・min^(-1);柱温:20℃;检测波长:272.321 nm,结果葛根素,龙胆苦苷、阿魏酸、蛇床子素、异欧前胡素分别在0.0366〜0.7328 mg・ml^(-1)(r=0.9994),0.0819〜1.6371 mg・ml^(-1)(r=0.9998),0.0027〜0.0532 mg・ml^(-1)(r=0.9999),0.0074〜0.14824 mg・ml^(-1)(r=0.9996),0.0033〜0.0656 mg・ml^(-1)(r=0.9997)范围内线性关系良好,平均加样回收率分别为101.54%、100.51%、94.56%、99.34%、97.94%,其RSD分别为1.82%.0.66%.1.79%.1.47%.1.93%。结论建立的多成分含量测定的方法可靠简便、重现性好,可用于温经活血搽剂的质量控制。
Objective To establish an HPLC-DAD method for simultaneous determination of contents of puerarin,gentiopicroside,ferulic acid,osthole and isoimperatorin in Wenjing Huoxue liniment.Methods Quantitative analysis was carried out on an Eclipse XDB-C_(18) column(4.6 mm×250 mm,5 pm)with a mobile phase composed of acetonitrile-0.1%phosphoric acid aqueous solution by gradient elution.The flow rate was 1.0 ml•min^(-1),the column temperature was maintained at 20℃,and the detection wavelengths were set at 272 and 321 nm.Results The linear ranges of puerarin,gentiopicroside,ferulic acid,osthole and isoimperatorin were 0,0366 to 0.7328 mg•ml^(-1)(r=0.9994),0,0819 to 1.6371 mg・ml^(-1)(r=0.9998),0.0027 to 0.0532 mg•ml^(-1)(r=0.9999),0.0074 to 0.14824 mg•ml^(-1)(r=0.9996)and 0.0033 to 0.0656 mg•ml^(-1)(r=0.9997),respectively.The average recoveries were 101,54%,100.51%,94.56%,99.34%and 97.94%,respectively while RSDs were 1.82%,0.66%,1.79%,1.47%and 1.93%,respectively.Conclusion The established method for determination of multi-component contents is reliable,simple and reproducible,which is suitable for quality control of Wenjing Huoxue liniment.
