摘要
目的研究大活络丸炼蜜中5-羟甲基糠醛的含量测定方法。方法采用高效液相色谱法,Agilent ZORBX SB-C18色谱柱(4.6 mm×250 mm,5μm),流动相为0.1%磷酸溶液-乙腈(95∶5),检测波长:284 nm,流速:1.0 ml·min^(-1),柱温:35℃。结果5-羟甲基糠醛在1.0~25.8μg·ml^(-1)有良好的线性关系,精密度良好,RSD<2.0%;加样回收率为100.5%,RSD为1.3%;7个厂家、28批大活络丸中的5-羟甲基糠醛含量为0.08~1.80 mg·g^(-1)。结论该方法准确可靠、重复性好,可用于测定大活络丸中5-羟甲基糠醛的含量。
Objective To establish a method for content determination of 5-hydroxymethyl furfural in refined honey of Dahuoluo pills.Methods HPLC was used.An Agilent ZORBX SB-C18 column(4.6 mm×250 mm,5μm)was selected for the separation of 5-hydroxymethyl furfural with 0.1%phosphoric acid and acetonitrile(95∶5)as the mobile phase at a rate of 1.0 ml·min^(-1).The detection wavelength was 284 nm,and the column temperature was 35℃.Results There was a good linear relationship within the range of 1.0 to 25.8μg·ml-1.The precision was satisfactory with relative standard deviations(RSD)less than 2.0%.The average recovery was 100.5%and RSD was 1.3%.The content of 5-hydroxymethyl furfural from 28 batches produced in 7 companies ranged from 0.08 to 1.80 mg·g-1.Conclusion This method for content determination of 5-hydroxymethyl furfural is accurate,reliable and repeatable,which can be used for determining the content of 5-hydroxymethyl furfural.
