摘要
从河北邯郸某虹鳟养殖场的底泥中分离得到一株革兰氏阴性菌,对其形态特征及16S rRNA基因序列进行测定分析并做生理生化鉴定,确定为一株托拉斯假单胞菌(Pseudomonas tolaasii),编号为G86。经发酵培养试验得到优化后的培养基和培养条件为:温度10℃,pH 6,转速180 r/min。以亚硝酸钠为唯一氮源,在上述培养条件下在72 h对总氮和亚硝酸盐氮的去除率分别为33.05%和31.85%。本次分离得到的托拉斯假单胞菌G86可以作为养殖生产上降解亚硝酸盐的备选菌株。
A gram-negative bacterium was isolated from the sediment of a rainbow trout farm in Handan,Hebei Province.Its morphological characteristics and 16S rRNA gene sequence were determined and analyzed,and its physiological and biochemical identification was carried out.It was identified as Pseudomonas tolasii,coded by G86.The optimized culture medium and conditions were obtained by fermentation trial:temperature 10℃,pH 6,rotation speed 180 rpm.With sodium nitrite as the sole nitrogen source,the removal rates of total nitrogen and nitrite nitrogen were 33.05%and 31.85%respectively at 72 h under the above culture conditions.The isolated Pseudomonas tolasii G86 can be used as an alternative strain for degradation of ammonia nitrogen in aquaculture.
