干扰脊椎蛋白2表达抑制肺腺癌细胞增殖和转移能力的研究 被引量:1

摘要 目的分析脊椎蛋白2(SPON2)在肺腺癌细胞中的表达及其对肺腺癌细胞增殖和转移的影响。方法2021年1月—2022年4月于沧州市中心医院进行实验,收集2015年1月—2016年12月医院胸外科行手术切除的肺腺癌组织及其配对的癌旁组织样本各86份,采用免疫组织化学法(IHC)检测肺腺癌组织和配对的癌旁组织中SPON2表达水平,并分析SPON2的表达与肺腺癌患者临床病理参数和预后的关系。培养肺腺癌细胞A549,分为si-NC组(转染NC siRNA)和si-SPON2组(转染SPON2 siRNA),Western-blot检测2组细胞中SPON2、细胞周期蛋白D1(Cyclin D1)、细胞周期蛋白依赖激酶4(CDK4)、基质金属蛋白酶7(MMP-7)、MMP-9的表达;CCK8实验和集落形成实验检测2组细胞的增殖能力;流式细胞仪检测2组细胞周期;Transwell实验检测2组细胞的转移能力。结果与癌旁组织比较,肺腺癌组织中SPON2阳性表达率显著上调(χ^(2)=5.821,P=0.016)。在肺腺癌T分期T3-4期、N分期N1-2期、TNM分期Ⅲ-Ⅳ期患者中SPON2表达分别高于T1-2期、N0期、Ⅰ-Ⅱ期(χ^(2)/P=7.481/0.006,5.331/0.021,9.139/0.003)。与低表达SPON2的肺腺癌患者比较,高表达SPON2的肺腺癌患者预后较差,差异具有统计学意义(χ^(2)=4.877,P=0.027)。si-SPON2组细胞中SPON2的表达明显高于si-NC组(t=28.343,P<0.001)。与si-NC组比较,si-SPON2组A549细胞的增殖和转移能力降低,细胞周期阻滞在G0/G1期,cyclinD1、CDK4、MMP-7、MMP-9表达减少,差异均有统计学意义(P<0.05)。结论SPON2在肺腺癌中表达上调,可能通过调控细胞周期相关蛋白及转移相关蛋白促进肺腺癌的增殖和转移能力,可能是治疗肺腺癌的潜在靶点。 Objective To analyze the expression of spinal protein 2(SPON2)in lung adenocarcinoma cells and its effect on the proliferation and metastasis of lung adenocarcinoma cells.Methods From January 2021 to April 2022,the experiment was carried out in Cangzhou Central Hospital.86 lung adenocarcinoma tissues and their paired adjacent tissue samples were collected from January 2015 to December 2016,respectively.Immunohistochemical method(IHC)was used to detect the expression level of SPON2 in lung adenocarcinoma tissues and paired adjacent tissues,and analyze the relationship between the expression of SPON2 and clinical pathological parameters and prognosis of lung adenocarcinoma patients.Lung adenocarcinoma cells A549 were cultured and divided into si NC group(transfected with NC siRNA)and si SPON2 group(transfected with SPON2 siRNA).Western blot was used to detect the expression of SPON2,cyclin D1,cyclin dependent kinase 4(CDK4),matrix metalloproteinase 7(MMP-7),and MMP-9 in the two groups;CCK8 test and colony formation test were used to detect the proliferation ability of the two groups of cells;The cell cycle was detected by flow cytometry;Transwell test was used to detect the metastatic ability of the two groups of cells.Results Compared with the adjacent tissues,the positive expression rate of SPON2 in lung adenocarcinoma tissues was significantly increased(χ^(2)=5.821,P=0.016).The expression of SPON2 in lung adenocarcinoma patients with T stage T3-4,N stage N1-2,TNM stageⅢ-Ⅳwas higher than that of T1-2,N0,Ⅰ-Ⅱ,respectively(χ^(2)/P=7.481/0.006,5.331/0.021,9.139/0.003).Compared with lung adenocarcinoma patients with low expression of SPON2,lung adenocarcinoma patients with high expression of SPON2 have a poor prognosis,and the difference is statistically significant(χ^(2)=4.877,P=0.027).The expression of SPON2 in si-SPON2 group was significantly higher than that in si-NC group(t=28.343,P<0.001).Compared with the si NC group,the proliferation and metastasis of A549 cells in the si SPON2 group decreased,the c
