β-榄香烯对胃癌细胞BGC-823细胞周期和血管生成相关蛋白的影响研究 被引量:6

摘要 目的研究β-榄香烯对胃癌细胞BGC-823细胞周期蛋白和血管生成相关蛋白的影响。方法以胃癌细胞BGC-823作为研究对象,实验分为对照组和50、100、200、400、800μmol/Lβ-榄香烯组,对照组中加入含有二甲亚砜的新鲜培养基,β-榄香烯处理组分别加入含有50、100、200、400、800μmol/Lβ-榄香烯的新鲜培养基。分别于干预后12、24、48 h,采用四甲基偶氮唑蓝(MTT)法检测细胞增殖能力,采用酶联免疫吸附试验(ELISA)检测细胞培养上清液中血管内皮生长因子(VEGF)、前列腺素E2(PEG2)水平。于干预后24 h,采用流式细胞术检测细胞凋亡情况,采用蛋白质印迹法检测细胞周期素D1(Cyclin D1)、细胞周期蛋白依赖激酶4(CDK4)、环氧化物酶2(COX-2)表达情况。结果干预12~48 h,所有分组细胞增殖均随时间延长显著降低,差异均有统计学意义(P<0.05);相同培养时间下,随着β-榄香烯浓度升高,BGC-823细胞增殖率呈显著降低趋势,差异均有统计学意义(P<0.05);相同β-榄香烯浓度下,随着培养时间延长,BGC-823细胞增殖率呈显著降低趋势,差异均有统计学意义(P<0.05)。对照组以及50、100、200、400、800μmol/Lβ-榄香烯浓度处理组BGC-823细胞培养24 h时的凋亡率分别为(5.47±0.62)%、(11.37±1.36)%、(18.92±2.17)%、(22.62±2.03)%、(33.27±3.27)%、(44.18±4.92)%,随着β-榄香烯浓度增加,细胞凋亡率呈显著增加趋势,差异均有统计学意义(P<0.05)。与对照组比较,200μmol/Lβ-榄香烯组和800μmol/Lβ-榄香烯组BGC-823细胞Cyclin D1、CDK4和COX-2表达水平均显著降低,且800μmol/Lβ-榄香烯组Cyclin D1、CDK4和COX-2表达水平均显著低于200μmol/Lβ-榄香烯组,差异均有统计学意义(P<0.05)。干预12~48 h,2个β-榄香烯组VEGF和PEG2表达随时间延长显著降低,差异均有统计学意义(P<0.05),而对照组不同时间点差异无统计学意义(P>0.05)。同一时间下,与对照组比较,200μmol/Lβ-榄香烯组和800μmol/Lβ-榄 Objective To study effects ofβ-elemene on cyclin and angiogenesis related proteins in gastric cancer cells BGC-823.Methods Gastric cancer cell BGC-823 was used as the research object,and the experiment was divided into control group and 50,100,200,400,800μmol/Lβ-elemene groups.The fresh medium containing dimethyl sulfoxide was added to the control group,and the fresh medium containing 50,100,200,400 and 800μmol/Lβ-elemene was added to theβ-elemene treatment group,respectively.At 12 h,24 h,and 48 h after the intervention,the cell proliferation was detected by MTT method,and the levels of vascular endothelial growth factor(VEGF),and prostaglandin E2(PEG2)in cell culture supernatant were detected by enzyme-linked immunosorbent assay(ELISA).At 24 h after intervention,flow cytometry was used to detect cell apoptosis,Western Blotting was used to detect the expression of cyclin-D1(Cyclin D1),cyclin-dependent kinase 4(CDK4)and cyclooxygenase 2(COX-2).Results Under the same culture time,the proliferation rates of BGC-823 cells decreased significantly with the increase ofβ-elemene concentration,the differences were statistically significant(P<0.05).Under the sameβ-elemene concentration,the proliferation rate of BGC-823 cells decreased significantly with the extension of culture time,the differences were statistically significant(P<0.05).The apoptosis rates of BGC-823 cells in control group and treatment groups with 50,100,200,400,800μmol/Lβ-elemene were(5.47±0.62)%,(11.37±1.36)%,(18.92±2.17)%,(22.62±2.03)%,(33.27±3.27)%,(44.18±4.92)%,respectively.Compared with the control group,the expression levels of Cyclin D1,CDK4 and COX-2 in BGC-823 cells in the 200μmol/Lβ-elemene group and 800μmol/Lβ-elemene group were significantly decreased,while the expression levels of Cyclin D1,CDK4 and COX-2 in the 800μmol/Lβ-elemene group were significantly lower than those in the 200μmol/Lβ-elemene group,and the differences were statistically significant(P<0.05).After 12-48 hours of intervention,the expressions of VEGF
