摘要
目的:建立柏参洗剂君药的鉴别和含量测定标准,用于控制和评价该制剂的质量。方法:采用薄层色谱法(TLC),摸索方中各药味特别是君药苦参、黄柏的鉴别方法,苦参使用硅胶G薄层板,展开剂为甲苯-丙酮-甲醇(8:3:0.5);黄柏采用硅胶G薄层板,正丁醇-冰醋酸-水(7:1:2)为展开剂;含量测定选择黄柏碱为指标成分,色谱条件为乙腈-0.1%磷酸水溶液(含2mg/mL的十二烷基磺酸钠),比例为36:64;流速1.0mL/min,检测波长284nm,柱温25℃,进样量10μL。结果:供试品中检出明显、清晰的苦参、黄柏对应斑点,阴性对照无干扰;10批供试品含量测定结果显示,黄柏碱的含量为0.6017~0.8065mg/mL。结论:本研究所建立的质量标准适宜、准确、高效,能够科学、规范地评价柏参洗剂的质量。
Objective:To establish the standard for the identification and content determination of Baishen lotion,and to control and evaluate the quality of the preparation.Methods:TLC was used to identify the ingredients of radix Sophora flavescens and Phellodendron phellodendron.Silica gel G thin layer plate was used for radix Sophora flavescens,and the developing agent was toluene-acetone-methanol(8:3:0.5).Silica gel G thin layer was used for Phellodendri chinensis with n-butanol-glacial acetic acid-water(7:1:2) as the developing agent.The chromatographic conditions were acetonitrile-0.1% phosphoric acid aqueous solution(containing 2mg/mL sodium dodecyl sulfonate),and the ratio was 36:64.The flow rate was 1.0 mL/min,the detection wavelength was 284nm,the column temperature was 25℃,and the injection volume was 10μL.Results:Obvious and clear corresponding spots of Sophora flavescens and Phellodendron phellodendri were detected in the test samples,and no interference was found in the negative control.The content determination results of 10 batches of samples showed that the content of phellodendrine was 0.6017~0.8065mg/mL.Conclusion:The quality standard established in this study is suitable,accurate and efficient,which can evaluate the quality of Baishen lotion scientifically and normatively.
