摘要
目的 比较可见、近红外、中红外三种光谱技术用于大黄炭炮制程度评价的可行性,为大黄炭及其他炭药的快速质量检测提供参考。方法 改变加热温度(180~320℃)和加热时间(10~60 min)制备不同炮制程度的大黄炭,结合颜色特征与成分含量将大黄炭按照炮制程度分组,分别使用可见、近红外、中红外三种光谱技术对不同组大黄炭进行鉴别分析。结果 可见光谱显示大黄炭炮制程度增加时对可见光的反射能力降低。基于近红外光谱的SIMCA分类模型对不同炮制程度的大黄炭鉴别准确度最高。中红外光谱能够解释大黄炭炮制程度增加时常量成分变化规律。结论 近红外光谱可以直接检测大黄切片,反映炭化程度增加时物理与化学性质的变化,可用于大黄炭炮制程度快速评价与炮制过程在线监测。中红外光谱可以直接检测大黄粉末,识别不同炮制程度大黄炭的常量成分特征,可作为大黄炭炮制终点鉴别标准的指标之一。
Objective To rapidly assess the quality of rhubarb charcoal and other carbon medicine by comparing the feasibility of visible,near-infrared (NIR) and mid-infrared (MIR) spectroscopic techniques in evaluating the processing degree of rhubarb charcoal.Methods Rhubarb charcoal samples processed to different degrees were prepared by altering the baking temperature (180~320℃) and time (10~60 min).These rhubarb charcoal were divided into several groups according to the processing degrees by the color and composition features.Visible,NIR and MIR spectroscopic techniques were used to discriminate the rhubarb charcoal with different processing degrees.Results With the increase of the processing degree,the reflectance of visible lights on the surface of rhubarb charcoal slices decreased.The NIR-based SIMCA model showed the best identification accuracy of rhubarb charcoal samples processed to different degrees.Bulk chemical changes of rhubarb charcoal during the process were revealed by MIR spectroscopy.Conclusion NIR spectroscopy can measure the rhubarb charcoal slices directly and indicate both the physical and chemical changes as the processing degree increases.NIR spectroscopy can be used in the rapid assessment of the processing degree and the on-line monitoring of the processing process of rhubarb charcoal.MIR spectroscopy can measure the rhubarb charcoal powders directly and assign the organic and inorganic characteristics of rhubarb charcoal with different processing degrees.It is probable to include MIR spectroscopy in the identification criterion for the processing endpoint of the rhubarb charcoal.
