余甘子果渣抑菌免洗凝胶的制备工艺研究 被引量:2

摘要 目的:测定余甘子果渣中的化学成分,优选提取工艺与免洗凝胶的制剂处方,开发余甘子果渣抑菌免洗凝胶。方法:采用高效液相色谱法(HPLC)测定余甘子果渣与余甘子药材中没食子酸、柯里拉京、诃黎勒酸与鞣花酸的含量;通过抑菌圈筛选余甘子果渣中主要抑菌活性成分筛选;采用单因素实验设计优化余甘子果渣的提取工艺;以感官评价及凝胶理化性质为指标,采用Box-Behnken响应面法优选余甘子果渣抑菌免洗凝胶处方。结果:余甘子果渣与余甘子药材的成分基本相同,没食子酸、柯里拉京、诃黎勒酸与鞣花酸在果渣的含量中约为药材中含量的21.99%~67.63%、0.52%~4.79%、7.38%~8.79%、10.15%~16.10%。诃黎勒酸、柯里拉京、没食子酸对白念珠菌、大肠杆菌和金黄色葡萄球菌的最低抑菌浓度(MIC)均低于10μg/mL,而鞣花酸的MIC为16.38~32.75μg/mL。以没食子酸、柯里拉京与诃黎勒酸为评价指标,得到最优提取工艺为取余甘子果渣,加入10倍体积的70%乙醇,回流2 h,提取1次。通过Box-Behnken响应面法,得到最优凝胶基质配方为30%余甘子果渣提取液、0.4%卡波姆U20、3%甘油、4%的95%乙醇、三乙醇胺调pH值到5~6,其余为纯化水。结论:余甘子果渣具有较好的开发价值,抑菌免洗凝胶的制备工艺可行,为中药固废物的综合开发利用提供了技术路径。 Objective:To determine the chemical composition of Phyllanthus emblica pomace and optimize the extraction process and formulation prescription of no-wash gel,thereby developing P.emblica pomace no-wash gel.Methods:High-performance liquid chromatography(HPLC)method was used to determine the content of gallic acid,corilagin,chebulagic acid,and ellagic acid in P.emblica pomace and Phyllanthi Fructus.The main antibacterial active components in P.emblica pomace were screened out by the inhibition zone.The single-factor experimental design was used to optimize the extraction process of P.emblica pomace.Taking sensory evaluation and physical and chemical properties of no-wash gel as indicators,the Box-Behnken response surface methodology was used to optimize the formulation of the bacteriostatic no-wash gel of P.emblica pomace.Results:The components of P.emblica pomace and Phyllanthi Fructus were basically the same.The content of gallic acid,corilagin,chebulagic acid,and ellagic acid in P.emblica pomace was about 21.99%~67.63%,0.52%~4.79%,7.38%~8.79%,and 10.15%~16.10% of the content in Phyllanthi Fructus.The minimal inhibitory concentration(MIC)of chebulagic acid,corilagin,and gallic acid against Monilia albican,Escherichia coli,and Staphylococcus aureus were all lower than 10μg/mL,while the MIC of ellagic acid was 16.38~32.75μg/mL.Taking gallic acid,corilagin,and chebulagic acid as evaluation indicators,the optimal extraction process was adding 10 times the volume of 70%ethanol in P.emblica pomace,refluxing for 2 h,and extracting it once.The optimized formula obtained by the Box-Behnken response surface method was 30%of raffinate residue extract,0.4% of carbomer U20,3% of glycerol,4% of 95%ethanol,triethanolamine with a pH value of 5~6,and purified water.Conclusion:P.emblica pomace has good development value,and the preparation process of no-wash gel is feasible,which provides a technical path for the comprehensive development and utilization of solid waste of Chinese medicine.
