室管膜下区神经前体细胞在衰老过程中的凋亡研究

摘要 目的:检测在小鼠衰老过程中室管膜下区(subventricular zone,SVZ)不同类型神经前体细胞和纹状体区神经元的凋亡程度。方法:1月龄、24月龄小鼠,灌注取脑、冰冻切片,行Nestin、胶质纤维酸性蛋白(glial fibrillary acidic protein,GFAP)、双皮质素(doublecortin,DCX)、NeuN分别与Cleaved caspase-3免疫荧光双标检测。结果:与1月龄小鼠相比,24月龄小鼠SVZ区Nestin阳性神经干细胞、GFAP阳性神经干细胞、DCX阳性成神经细胞和纹状体区NeuN阳性神经元的凋亡比例均增加,但在凋亡程度上有差别。SVZ区DCX阳性成神经细胞的凋亡程度最高,GFAP阳性神经干细胞的凋亡程度最低。结论:24月龄小鼠SVZ区神经前体细胞凋亡程度增加,可能在大脑衰老中起着重要的作用。 Objective:To detect the apoptosis of different types of neural precursor cells in subventricular zone(SVZ)and neurons in striatum during aging in mice.Methods:Mice aged 1 month and 24 months were perfused,and the brains and frozen sections were taken.Nestin,glial fibrillary acidic protein(GFAP),doublecortin(DCX)and NeuN/Cleaved caspase-3 were detected by immunofluorescence double labeling.Results:Compared with 1-month-old mice,the apoptosis proportion of nestin positive neural stem cells,GFAP positive neural stem cells,DCX positive neuroblasts and NeuN positive neurons in striatum increased in 24-month-old mice,but there were differences in the degree of apoptosis.The apoptosis degree of DCX positive neuroblasts in SVZ was the highest,and that of GFAP positive neural stem cells was the lowest.Conclusion:The degree of apoptosis of neural precursor cells in SVZ area of 24-month-old mice increases,which may play an important role in brain aging.
