摘要
目的评估2,4-二硝基苯酚(DNP)是否具有辐射增敏的能力。方法采用MTT法及细胞克隆形成实验检测不同浓度DNP对Hela、KB细胞增殖能力的影响;细胞克隆形成实验检测DNP对Hela、KB细胞的辐射增敏作用;流式细胞仪检测不同条件下Hela、KB细胞周期及线粒体膜电位变化,并利用透射电子显微镜观察自噬小体形成;western blotting检测自噬相关蛋白LC-Ⅰ、LC-Ⅱ的表达。结果DNP浓度越高对Hela、KB细胞的增殖抑制越明显;在200μmol/L的DNP作用下,Hela、KB细胞的SERD0分别为1.45、1.18;Hela、KB细胞经过4 Gy X线照射并联合200μmol/L DNP作用24 h后,G_(2)/M期细胞明显增多,线粒体膜电位显著降低;DNP能够促进Hela、KB细胞自噬小体形成;在DNP作用后Hela及KB细胞的LC-Ⅱ表达明显升高,与对照组相比LC-Ⅱ/LC-Ⅰ值也明显有所升高。结论DNP能够以浓度依赖的方式抑制肿瘤细胞增殖,并且具有辐射增敏作用。DNP可能是一种潜在的辐射增敏剂。
Objective To assess whether 2,4-dinitrophenol(DNP)has the ability of radio sensitization.Methods The MTT method and cell clone formation experiment were used to detect the effect of different concentrations of DNP on the proliferation of Hela and KB cells.The cell clone formation experiment was also used to detect the radio sensitizing effect of DNP on Hela and KB cells.Flow cytometry was used to detect the change of the Hela and KB cells cycle and mitochondrial membrane potential in different conditions,and the transmission electron microscope was used to observe the formation of the autophagosome.The expressing of autophagy-related proteins LC-Ⅰ,LC-Ⅱwas detected by the western blotting.Results With the concentration of DNP increased,the proliferation of Hela and KB cells was more obviously inhibited.When the concentration of DNP was 200μmol/L,the SERD0 of Hela and KB cells were 1.45 and 1.18.After Hela and KB cells were irradiated by 4 Gy X-ray combined with 200μmol/L DNP for 24 h,the number of G_(2)/M cells increased significantly and the mitochondrial membrane potential decreased significantly.DNP could promote the formation of autophagosome of Hela and KB cells.After DNP treatment,the expression of LC-Ⅱin Hela and KB cells was significantly increased,and the ratio of LC-Ⅱ/LC-Ⅰwas also significantly higher than that in the control group.Conclusion DNP can inhibit the proliferation of tumor cells in a concentration-dependent manner and has radio sensitizing effect.DNP may be a potential radiosensitizer.
