摘要
目的观察二甲双胍对糖尿病合并抑郁症大鼠抑郁行为的影响,并探讨可能机制。方法45只大鼠中随机抽取10只设为对照组,其余35只建立糖尿病合并抑郁症大鼠模型,30只建模成功,随机分为模型组、实验A组、实验B组,各10只。实验A、B组分别腹腔注射二甲双胍150 mg·kg^(-1)、300 mg·kg^(-1),对照组、模型组腹腔注射等体积生理盐水。采用旷场实验检测大鼠自发活动度;采用糖水消耗实验检测大鼠糖水偏嗜度;采用放射免疫分析法检测下丘脑-垂体-肾上腺轴(HPA)轴活性;采用免疫印迹法检测海马组织闭锁小带蛋白-1(ZO-1)蛋白表达。结果5 min内总移动距离、糖水偏嗜度、血清皮质醇(CORT)水平、海马组织ZO-1蛋白表达量指标,模型组与对照组比较,实验A组与模型组比较,实验B组与实验A组比较,差异均有统计学意义(P<0.05)。结论二甲双胍可改善糖尿病合并抑郁症大鼠抑郁样行为,减轻血脑屏障损伤,可能通过抑制下丘脑-垂体-肾上腺轴(HPA)轴活性来发挥作用。
Objective To observe the effects of metformin on the depressive behaviors in rats with diabetes and depression syndrome,and explore its possible mechanism.Methods Ten of 45 rats were randomly selected as the control group.The remaining 35 rats were modeled with diabetes complicated with depression.Thirty rats were successfully modeled and randomly divided into a model group,an experimental-A group,and an experimental-B group,with 10 rats in each group.The experimental-A group and experimental-B group were given intraperitoneal injections of metformin 150 and 300 mg·kg^(-1),respectively.The control group and model group were given intraperitoneal injections of equal volume of normal saline.The spontaneous activity of rats were measured by the open field experiment.The sugar water preference of rats were detected by the sugar water consumption experiment.The activity of the hypothalamic-pituitary-adrenal axis (HPA axis) was measured by radioimmunoassay.The protein expression of zonule atresia protein-1 (ZO-1) in hippocampus of rats was detected by Western blotting.Results There were statistically signi?cant di?erences in the total movement distance within 5 minutes,sugar water preference,serum cortisol (CORT) level,and hippocampal ZO-1 protein expression between the model group and control group,the experimental-A group and model group,the experimental-B group and experimental-A group (P<0.05).Conclusion Metformin can ameliorate the depressive-like behaviors in rats with diabetes complicated by depression and reduce the blood-brain barrier damage,possibly by inhibiting the activity of the hypothalamic-pituitary-adrenal axis.
