摘要
目的:建立同时检测牡丹皮中芍药苷、丹皮酚含量的测定方法。方法:采用高效液相双波长测定法,流动相为乙腈-0.1%磷酸溶液,检测波长为232 nm和274 nm,流速为1.0 mL/min,进样量10 μL,色谱柱为安捷伦Eclipse Plus C18(250 mm×4.6 mm,5 μm),柱温25 ℃。结果:芍药苷和丹皮酚分别在0.000 6~0.291 9 mg/mL;0.000 6~0.300 6 mg/mL浓度范围内线性关系良好;精密度、稳定性、重复性相对标准偏差均小于2.0%;加样回收率均值分别为101.50%、99.40%。结论:牡丹皮中芍药苷和丹皮酚含量在不同产区有很大的差异。此法方便快捷,准确可靠,能够为牡丹皮的品控提供保障。
Objective:To establish a method that can detect the content of paeoniflorin and paeonol in moutan cortex simultaneously.Methods:High-performance liquid phase dual-wavelength determination method was used.It was gradient elution with the water-0.1% phosphoric acid.The 232 nm and 274 nm were as its detection wavelengths.The flow rate was 1.0 mL/min.The injection volume was 10 μL and the chromatographic column was Agilent Eclipse Plus C18 (250 mm×4.6 mm,5 μm).The column temperature was 25 °C.Results:The paeoniflorin and paeonol has good linear relationship in the range of 0.000 6~0.291 9 mg/mL and 0.000 6~0.300 6 mg/mL.The relative standard deviation of precision,stability and repeatability were less than 2.0%.The recovery rates of these two components were 101.50% and 99.40% respectively.Conclusion:The contents of paeoniflorin and paeonol in moutan cortex are differences vary greatly in different producing areas.This method is convenient,quick,accurate and reliable,and can effectively control the quality of moutan cortex.
