摘要
目的建立RP-HPLC同时测定丹膝颗粒中的丹酚酸B、丹皮酚和丹参素含量的分析方法。方法供试品加甲醇经超声提取。采用Agilent Zorbax eclipse SB-C18色谱柱(250 mm×4.6 mm,5μm)对提取液进行分离,以0.1%甲酸水溶液-乙腈为流动相,梯度洗脱,流速1.0 ml/min,柱温30℃,检测波长282 nm。结果丹酚酸B、丹皮酚和丹参素在2.0~100μg/ml范围内,与峰面积线性关系良好(r均大于0.995);检测限分别为0.12,0.08,0.28μg/ml;加样回收率分别为95.3%,93.2%,100.7%;精密度和重复性(n=6)RSD均在5.0%以内。结论本方法具有前处理简单、检出限低、重复性好、检测结果准确等优点,可以用于丹膝颗粒的质量控制。
Objective To establish a RP-HPLC method for simultaneous determination of salvianolic acid B,paeonol and tanshinol in Danxi Granules.Methods The sample was extracted by ultrasonic treatment with methanol.Agilent Zorbax eclipse SB-C18(250 mm×4.6 mm,5μm)column was adopted for separation of the extract.Using 0.1%formic acid aqueous solution-acetonitrile as mobile phase,gradient elution was applied.The flow rate was 1.0 ml/min,the column temperature was 30℃and the detection wavelength was 282 nm.Results Salvianolic acid B,paeonol and tanshinol showed good linear relationship with peak area in the range 2.0-100μg/ml(r>0.995).Detection limits of salvianolic acid B,paeonol and tanshinol were 0.12,0.08 and 0.28μg/ml,respectively.The average recoveries were 90.0%-110.0%.RSDs of precision and repeatability test were within 5.0%(n=6).Conclusion The method has the advantages of simple pretreatment,low detection limit,good repeatability and accurate detection results,and can be used for quality control of Danxi Granules.
