SOCS6对前列腺癌分化的影响及其临床意义

摘要 目的研究SOCS6的表达情况对前列腺癌动物模型移植瘤形成的影响,分析其表达情况对前列腺肿瘤分化的影响,探讨其作用机制和临床意义。方法在人前列腺癌组织中,通过免疫组化检测SOCS6的表达情况并确定其主要表达部位。使用慢病毒载体构建稳定过表达SOCS6的人类前列腺癌PC3和LNCaP细胞株(pre-SOCS6),通过Western blot与空载组(pre-000)和正常细胞组(Normal)进行比较,对过表达组细胞进行鉴定。将SOCS6过表达组与空载组的两种前列腺癌细胞株PC3和LNCaP进行裸鼠肾包膜下接种,以形成动物移植瘤模型(PC3-pre-000、PC3-pre-SOCS6、LNCaP-pre-000、LNCaP-pre-SOCS6),并观察各组裸鼠成瘤的状况。取瘤内组织进行HE染色后镜下观察,分析SOCS6过表达后对前列腺肿瘤分化程度的影响。结果免疫组化显示SOCS6在前列腺癌样本组织中染色强度主要为中、弱阳性,且主要表达在前列腺上皮基底细胞层。Western blot实验结果显示SOCS6过表达组细胞SOCS6的蛋白表达水平显著高于正常组和空白组。通过HE染色观察裸鼠肿瘤组织可发现,上调前列腺癌中SOCS6的表达水平可促进肿瘤分化,降低肿瘤恶性程度。结论SOCS6在前列腺癌组织的表达水平与肿瘤分化程度密切相关,提示通过检测前列腺癌组织中SOCS6的表达,有助于区分低度恶性和高度恶性的前列腺癌,其可能作为预后判断的标记物和潜在的治疗靶标。 Objective To study the effect of SOCS6 expression on graft tumor formation in animal models of prostate cancer and further analyze the effect of its expression on prostate tumor differentiationand explore its mechanisms of action and clinical significance.Methods The expression of SOCS6 was examined by immunohistochemistry staining in human prostate cancer tissues and its major sites of expression were identified.Human prostate cancer PC3 and LNCaP cell lines stably overexpressing SOCS6 were constructed using a lentiviral vector(pre-SOCS6),and identification of cells in the overexpression group by Western blot compared to the null(pre-000)and normal cell groups(normal).Two prostate cancer cell lines,PC3 and LNCaP,from the SOCS6 stable overexpression group and the control null group,were inoculated subperitoneally in nude mice to form animal transplantation tumour models(PC3-pre-000、PC3-pre-SOCS6、LNCaP-pre-000、LNCaP-pre-SOCS6).Tumor formation was observed in each group of nude mice.Intratumoral tissues were taken for microscopic observation after HE staining to analyze the effect of SOCS6 overexpression on the degree of differentiation of prostate tumors.Results The intensity of immunohistochemical staining for SOCS6 gene in prostate cancer samples was mainly moderate/weakly positive,and it was mainly expressed in the basal cell layer of the prostate epithelium.The mRNA and protein expressionsof SOCS6 were higher in the SOCS6 overexpression group compared with the control group.The up-regulation of SOCS6 expression in prostate cancer could promote tumor differentiation and reduce tumor malignancy as observed by HE staining in nude mice tumor tissues.Conclusion The expression level of SOCS6 in prostate cancer tissues is closely related to the degree of tumor differentiation,suggesting that SOCS6 expression in prostate cancer tissuescouldhelp distinguish between low-grade malignant and highly malignant prostate cancers.Thus,SOCS6 might be used as a marker for prognosis and a potential therapeutic target of pro
