柴朴汤含药血清对气道上皮细胞株中MAL、CADM1、TSLP、YKL-40表达的影响 被引量:1

Effect of Chai Pu Decoction-containing serum on the expression of MAL,CADM1,TSLP,and YKL-40 in airway epithelial cells lines in vitro

摘要 目的探讨柴朴汤含药血清对气道上皮细胞(Airway epithelial cell,AEC)中T细胞成熟相关蛋白(MAL)、细胞黏附分子1(CADM1)、胸腺间质淋巴细胞生成素(TSLP)、甲壳质酶蛋白40(YKL-40)表达的影响。方法制备空白、肝郁型哮喘及柴朴汤含药血清,分别加入正常小鼠AEC中培养,分为正常组(A)、肝郁型哮喘组(B)和柴朴汤组(C)。采用酶联免疫吸附法(ELISA)检测TSLP、YKL-40在小鼠气道上皮细胞的上清液中的浓度,实时荧光定量PCR(qRT-PCR)法和蛋白免疫印迹法(WB)检测各组气道上皮细胞中MAL、CADM1、TSLP、YKL-40 mRNA及蛋白的表达情况。结果经肝郁型哮喘血清诱导后,气道上皮细胞中MAL、CADM1 mRNA及蛋白表达减少,TSLP、YKL-40 mRNA及蛋白表达增加,上清液中TSLP、YKL-40的浓度增加,与正常组相比有统计学意义(P<0.05);柴朴汤含药血清诱导后,气道上皮细胞中MAL mRNA和蛋白表达增加,TSLP、YKL-40 mRNA及蛋白表达减少,上清液中TSLP、YKL-40的浓度减少,与肝郁型哮喘组相比有统计学意义(P<0.05),CADM1 mRNA和蛋白表达无明显变化。结论①肝郁型哮喘血清可诱导AEC中CADM1、MAL表达降低,TSLP、YKL-40表达升高。②柴朴汤含药血清可上调肝郁型哮喘血清培养AEC中MAL的表达,下调TSLP、YKL-40表达。 Objective To investigate the effects of Chai Pu Decoction-containing serum on the expression of T-cell maturation-associated protein(MAL),cell adhesion molecule 1(CADM1),thymic mesenchymal lymphopoietin(TSLP)and chitinase protein 40(YKL-40)in airway epithelial cells.Methods Blank,liver-depression asthma and Chai Pu Decoction-containing sera were prepared and added to the airway epithelial cells of normal mice for culture,which were divided into normal group(A),liver-depression asthma group(B)and Chai Pu Decoction group(C).The expression of MAL,CADM1,TSLP,YKL-40 mR⁃NA and protein in each group were detected by quantitative real-time fluorescence PCR(qRT-PCR)and protein immunoblot⁃ting(WB).Results After serum induction of liver-depression asthma,MAL and CADM1 mRNA and protein expression de⁃creased in airway epithelial cells,TSLP and YKL-40 mRNA and protein expression increased,and the concentrations of TSLP and YKL-40 in the supernatant increased;after serum induction of Chai Pu Decoction containing medicine,MAL mRNA and protein expression increased,TSLP and YKL-40 mRNA and protein expression decreased,and the concentrations of TSLP and YKL-40 in the supernatant decreased,which was statistically significant compared with the liver-depressed asthma group(P<0.05),and CADM1 mRNA and protein expression did not change significantly.Conclusion①Liver-depressed asthma serum induced decreased expression of CADM1 and MAL and increased expression of TSLP and YKL-40 in AEC.②Chai Pu Decoction-containing serum up-regulated MAL expression and down-regulated TSLP and YKL-40 expression in AEC cultured with liv⁃er-depressed asthma serum.
