武夷山洋庄茶区水仙茶树丛枝菌根真菌多样性调查

An Investigation on Diversity of Arbuscular Mycorrhizal Fungi in the Rhizosphere of Shuixian Tea Tree in Yangzhuang Tea Area of Wuyishan City

摘要 为调查武夷山市洋庄茶区水仙茶树丛枝真菌(AMF)侵染特征及AMF多样性,采用醋酸-墨水法测定水仙茶树AMF侵染率,应用湿筛-倾斜蔗糖离心法对根际土壤中的AMF进行分离鉴定。结果表明:水仙茶树AMF侵染率在12%~100%,平均侵染率达68.4%,土壤类型对茶树根系的AMF侵染有一定影响,老丛水仙与一般水仙的AMF侵染率无显著差异(P﹥0.05)。武夷山市洋庄茶区茶树AMF资源较为丰富,从各样地根际土中共鉴定出3属26种AMF,其中球囊霉属16种,无梗囊霉属9种,盾巨孢囊霉属1种;聚丛球囊霉、明球囊霉、宽柄球囊霉、木薯球囊霉、粘屑球囊霉和光壁无梗囊霉的分布频度均大于50%,是常见种,其中光壁无梗囊霉的分布频度89.3%,为优势种。不同采样地茶树根际的AMF物种丰度(SR)表现出较大的差异。AMF群落Sφrenson相似性分析表明,水仙茶树根际土壤AMF群落构成与土壤类型有关,而与茶树生长年龄无关;老丛水仙与一般水仙茶树在AMF侵染率和AMF群落构成方面均无显著差异,说明两者茶品质上的差异与AMF无关。 To determine the infection characteristics and diversity of Arbuscular mycorrhizal fungi(AMF) in the rhizosphere of Carmellia sinensis CV. Fujian Shuixian in Yangzhuang tea area of Wuyishan City in Fujian Province, the AMF infection rate and the series of infection were measured by using an improved acetic acid ink staining method. The AMF species were isolated and identified by wet sieving sieve-inclined sucrose centrifugation method and microscopic observation method. The results showed that the AMF infection rate of all tested root samples ranged from 12% to 100%,with an average of 68.4%. The type of rhizosphere soil had a certain influence on AMF infection of tea plant root. There was no significant difference in root AM infection rate between old aged Shuixian tea tree and young Shuixian tea tree. There were abundant tea AMT resources in Yangzhuang tea area of Wuyishan City in Fujian Province. A total of 26 species belonging to 3 genera of AMF were isolated and identified from tested soil samples. There were 16 species in Glomus, 9 species in Acaulospora, 1 species in Scutellospra. Among them, Glomus aggregatum, G. clarum, G. magnicaule, G. manihotis, G. spurcum and Acaulospora laevis were the common species because their distribution frequence was all bigger than 50%. Acaulospora laevis was considered as the dominant species with a distribution frequence up to 89.3%. There was a significant difference in AMF species richness of rhizosphere soil between different tested plots. The analysis of Sφrenson similarity coefficient indicated that the communities of AMF had a positive correlation with the type of rhizosphere soil, and had no correlation with the growing age of tea tree. There was no significant difference in both of AM infection rate and AMF communities between the old aged Shuixian tea tree and young Shuixian tea tree, indicating that the difference in tea quality had nothing to do with the AMF in two kinds Shuixian tea trees.
