摘要
目的探讨通窍活血汤(TQHX)对兔视网膜分支静脉阻塞(BRVO)模型视网膜组织中骨桥蛋白(OPN)、白细胞介素-6(IL-6)、白细胞介素-8(IL-8),以及玻璃体中IL-6、IL-8表达的影响。方法30只新西兰白兔随机分为对照组(CG)、模型组(MG)、通窍活血汤组(TQHX),每组10只。CG组进行假手术,MG组、TQHX组兔双眼采用532 nm激光建立BRVO模型。造模成功后,TQHX组给予5.75 g/kg的TQHX灌胃,CG、MG组给予等体积蒸馏水灌胃,给药21 d后取兔双眼视网膜组织以及玻璃体,Western Blot法检测OPN和IL-6、IL-8在视网膜中的表达,酶联免疫吸附法(ELISA)检测IL-6、IL-8在玻璃体中的表达。结果(1)OPN:与CG组比较,MG组兔视网膜OPN表达升高(t=-6.903,P=0.000);与MG组比较,TQHX组兔视网膜OPN表达降低(t=3.479,P=0.003),差异均有统计学意义。(2)IL-6:与CG组比较,MG组兔视网膜和玻璃体IL-6表达升高(t_(视网膜)=-8.006,t_(玻璃体)=-4.725,均P=0.000);与MG组比较,TQHX组兔视网膜和玻璃体IL-6表达降低(t_(视网膜)=6.971,P=0.000;t_(玻璃体)=3.380,P=0.001),差异均有统计学意义。(3)IL-8:与CG组比较,MG组兔视网膜和玻璃体IL-8表达升高(t_(视网膜)=-8.908,P=0.000;t_(玻璃体)=-3.899,P=0.001);与MG组比较,TQHX组兔视网膜和玻璃体IL-8表达降低(t_(视网膜)=5.097,P=0.000;t_(玻璃体)=2.328,P=0.032),差异均有统计学意义。结论TQHX对BRVO模型兔视网膜组织中OPN、IL-6、IL-8以及玻璃体中IL-6、IL-8的表达水平具有下调作用,其可能对临床治疗BRVO或其他缺血缺氧性视网膜病变具有一定作用。
OBJECTIVE To investigate the effect of Tongqiao Huoxue Decoction(TQHX)on osteopontin(OPN),interleukin-6(IL-6),interleukin-8(IL-8)in the retina and vitreous body of branch retinal vein occlusion(BRVO)rabbit model.METHODS Thirty New Zealand white rabbits were randomly divided into three groups as control group(CG),model group(MG)and TQHX group with 10 rabbits in each group.Rabbits in CG underwent sham operation,subjects in MG and TQHX were irradiated with 532 nm laser to establish BRVO models.After successful modeling,rabbits in TQHX were given 5.75 g/kg TQHX by gavage,these in CG and MG were given equal volume distilled water by gavage,and after 21 d of feeding,the retinal tissues and vitreous body of both eyes were collected by anesthesia.OPN,IL-6 and IL-8 in the retinal tissues were detected by Western Blot.Elisa was used to detect the expression of IL-6 and IL-8 in rabbit vitreous body.RESULTS(1)OPN:Compared with CG,the expression of OPN increased in MG(t=-6.903,P=0.000);Compared with MG,the expression of OPN decreased in TQHX(t=3.479,P=0.003),and the differences were statistically significant.(2)IL-6:Compared with CG,the expression of IL-6 in retina and vitreous body of MG was increased(t_(reti)=-8.006,t_(vitr)=-4.725,all P=0.000);Compared with MG,the expression of IL-6 in retina and vitreous body of TQHX was decreased(t_(reti)=6.971,P=0.000;t_(vitr)=3.380,P=0.001),and the differences were statistically significant.(3)IL-8:Compared with CG,the expression of IL-8 in retina and vitreous body of MG was increased(t_(reti)=-8.908,P=0.000;t_(vitr)=-3.899,P=0.001);Compared with MG,the expression of IL-8 in retina and vitreous body of TQHX was decreased(t_(reti)=5.097,P=0.000;t_(vitr)=2.328,P=0.032),and the differences were statistically significant.CONCLUSIONS The TQHX showed down-regulating effects on the expression of OPN,IL-6,IL-8 in retinal and IL-6 and IL-8 in vitreous body of rabbit BRVO model.Therefore,it may have certain effects on BRVO or other ischemic and hypoxic retinopathy in clinic.
