摘要
药用植物愈伤组织诱导率及生长状态会直接影响其增殖及活性成分积累。为有效提高黄芩愈伤组织的诱导率、优化其生长条件,以黄芩组培苗的根、茎段和叶片为材料,经不同浓度的6-BA、NAA和2,4-D单因素、双因素以及L(933)正交试验处理,以黄芩愈伤组织诱导率和生长状态为评价指标,筛选其最佳外植体和培养基配方。结果发现,在单因素试验中,当6-BA质量浓度为1.0、1.5、2.0 mg/L时,有且只有茎段诱导状态较好,呈淡黄色,分化状态相对较低,最大诱导率可达88%;当NAA质量浓度为0.5、1.0、1.5 mg/L时,同样茎段诱导情况良好,分化较低,增殖快,生物量大,最大诱导率为72%;在2,4-D测试质量浓度范围内,愈伤组织呈乳白色至淡黄色,诱导率大小表现为茎段>根>叶片,且在0.1 mg/L时,茎段有最高诱导率(97%);故选择1.0、1.5、2.0 mg/L的6-BA,0.5、1.0、1.5 mg/L的NAA和0.05、0.15、0.25 mg/L的2,4-D进行双因素和正交试验,并确定茎段为愈伤组织诱导的最佳外植体。在6-BA和NAA组合试验中,发现不同浓度组合均发生分化现象,说明6-BA和NAA组合适合于芽的增殖培养。进一步通过正交试验结果分析,发现适合黄芩茎段愈伤组织诱导的最佳培养基为MS+1.0 mg/L 6-BA+1.0 mg/L NAA+0.25 mg/L 2,4-D,其诱导率高达94.16%,且愈伤组织颜色呈淡黄色,结构疏松,生长状态良好,可用于继代和增殖培养。
The callus induction rate and growth states of medicinal plants will directly affect their proliferation and accumulation of active components.To effectively improve the callus induction rate of Scutellaria baicalensis and optimize its growth conditions,in this study,taking roots,stem segments,and leaves of S.baicalensis as the materials,single factor,double-factor,and L9(33)orthogonal experiment treatments of different concentreations of 6-BA,NAA and 2,4-D were used to screen the optimal explant and medium for callus induction of S.baicalensis based on callus induction rate and growth states of S.baicalensis.The results showed that in the single factor test,when the concentration of 6-BA was 1.0,1.5,2.0 mg/L,only the stem segments had good induction state,its colour was pale yellow,differentiation state was low,and the maximum induction rate reached 88%.When the concentration of NAA was 0.5,1.0,1.5 mg/L,the stem segments also had good induction state with low differentiation,fast proliferation,and large biomass,the maximum induction rate was 72%.Within the range of test weight concentration of 2,4-D,the induced callus was milky white to pale yellow,induction rate of the explants was as the order of stem segments>roots>leaves,and the stem segments had the maximum induction rate of 97%at the concentration of 0.1 mg/L.Therefore,1.0,1.5,2.0 mg/L of 6-BA,0.5,1.0,1.5 mg/L of NAA,and 0.05,0.15,0.25 mg/L of 2,4-D were subjected to do two-factor and orthogonal experiments.And the stem segments were determined as the optimal explants for callus induction.The differentiation phenomenon existed in different concentration combinations in all two-factors experiments of 6-BA and NAA,indicating that the combination of 6-BA and NAA was suitable for the proliferation and culture of buds.The further results of orthogonal experiments showed that the optimal medium for callus induction from stem segments of S.baicalensis was MS+1.0 mg/L of 6-BA+1.0 mg/L of NAA+0.25 mg/L of 2,4-D with the highest induction rate of 94.16%and the callu
