摘要
以附加2%蔗糖的MS为基本培养基,添加不同的植物激素,进行滇黄芩体胚诱导与植株再生。结果表明:叶柄及不带腋茎段在含NAA0.2~1mg/L的培养基中可诱导愈伤组织;茎尖及带腋茎段在6-BA(或KT)0.5mg/L+IBA0.5mg/L培养基中诱导胚状体和芽丛,在6-BA(或KT)0.05mg/L+IBA 0.1mg/L培养基中增殖小植株;在1/2MS+IAA 1mg/L+3%蔗糖培养基中能产生带2~3级根的壮根;所有培养需附加1%琼脂粉,克服玻璃化现象。
Somatic embryo induction and plantlet regeneration of SScutellaria amoena C. H. Wright on MS medium supplemented with 2% sucrose and different the plant homones.The results showed: (i)calli were induced in the medium‘MS + NAA 0.2 - 1 mg/L' using leaf stalk and stem without axillary buds, (ii) embryoid and shonts were induced in the medium‘MS + 6-BA(或KT)0.5 mg/L + 1BA0.5 mg/L' and seedling multiplied in the medium ‘MS + 6-BA(或KT)0.05 mg/L + IBA0.1 mg/L' using stem apex and stem with axillary buds, (iii)roots with the second or third order were induced in the medium‘1/2 MS + IAA 1 mg/L' adding 3 % sucrose, (iv)in order to eliminate vitrification, all the medium have to add 1% agar.
