摘要
3-磷酸肌苷依赖性蛋白激酶1(PDK1)是一种高度保守的真核细胞激酶,是许多AGC激酶亚家族成员的核心调控因子。通过其对AGC激酶的调控进而控制许多基本细胞过程。本研究克隆获得了水曲柳PDK1的基因序列全长,将其命名为Fm PDK1(已上传至NCBI基因库,NCBI序列号:MG356659)。水曲柳Fm PDK1全长1470 bp,编码481个氨基酸;该基因内部有完整的open reading frame(ORF),存在螺旋-环-螺旋结构;亲水性不稳定蛋白;不存在信号肽,具有跨膜能力;亚细胞定位预测该蛋白最可能定位于细胞核内;水曲柳Fm PDK1与芝麻(Sesamum indicum XP_011094484.1)亲缘关系最近,同源性为83.9%;4℃低温处理下,Fm PDK1的表达量发生显著变化;100 mmol/L NaCl处理下,Fm PDK1的表达量也发生了变化但是并不显著,脱落酸(ABA)和赤霉素(GA)以及IAA(生长素)三种激素诱导下,Fm PDK1表达量均变化显著。水曲柳Fm PDK1对寒冷、盐等非生物胁迫以及激素信号做出了应答,有望为进一步研究水曲柳AGC激酶亚家族成员中PDK1基因的特性及功能提供科学依据。
Inosine 3-phosphate-dependent protein kinase 1(PDK1)is a highly conserved eukaryotic kinase and a core regulator of many AGC kinase subfamily members.Through its regulation of AGC kinase,it controls many basic cellular processes.In this study,the full-length gene sequence of Fraxinus mandshurica PDK1 was cloned and named Fm PDK1(uploaded to NCBI gene bank,NCBI serial number:MG356659).Fraxinus mandshurica FmPDK1 has a full length of 1470 bp,which encodes 481 amino acids;the gene has a complete open reading frame(ORF)with a helix-loop-helix structure;hydrophilic labile protein;no signal peptide,with transmembrane ability;Subcellular location predicts that the protein is most likely to be located in the nucleus;Fraxinus mandshurica Fm PDK1(Sesamum indicum XP_011094484.1)has the closest genetic relationship with a homology of 83.9%;under 4℃ low temperature treatment,the expression of Fm PDK1 changes significantly;Under the treatment of100 mmol/L NaCl,the expression of Fm PDK1 also changed but it was not significant.Under the induction of the three hormones of abscisic acid(ABA),gibberellin(GA)and IAA(auxin),the expression of Fm PDK1 changes significantly.Fraxinus mandshurica Fm PDK1 responds to cold,salt and other abiotic stresses and hormone signals.It is expected to lay the foundation for further research on the characteristics and functions of PDK1 genes in members of the Fraxinus mandshurica AGC kinase subfamily.
