摘要
目的研究金银花的化学成分及其抗菌消炎活性。方法采用多种色谱方法对金银花的化学成分进行分离纯化,利用光谱法对化合物进行结构鉴定;采用抑制一氧化氮(NO)生成实验、抑制环氧化酶活性实验评价化合物的抗炎活性;采用微量稀释法对化合物进行常规菌、厌氧菌抑制活性评价。结果从金银花药材中分离并鉴定了22个主要化学成分,分别为断氧马钱子苷(1)、secologanol(2)、金吉苷(3)、8-表金吉苷(4)、獐牙菜苷(5)、7-酮马钱素(6)、断氧马钱子苷半缩醛内酯(7)、表断氧马钱子苷半缩醛内酯(8)、threoninosecologanin(9)、5S-羧基异胡豆苷(10)、坎卡酯苷(11)、1,2-O-二咖啡酰基-3-羟基-环戊醇(12)、4,5-O-二咖啡酰基奎宁酸(13)、咖啡酸(14)、绿原酸(15)、紫丁香苷(16)、芦丁(17)、supinaionodie B(18)、cordysinin B(19)、鸟嘌呤核苷(20)、腺嘌呤核苷(21)、烟酰胺(22)。活性筛选结果表明分离得到的化合物并没有明显的NO生成抑制活性,也不能抑制环氧化酶1和2的活性;化合物2、5、11、19对厌氧菌变异链球菌显示出抑制作用,化合物4、6、8、15对金黄色葡萄球菌显示出一定的抑制作用。结论本研究从金银花药材中分离鉴定了22个单体化合物,其中化合物11、12、18、22为首次从该种植物中分离得到,化合物2、4、5、6、8、11、15、19具有抗菌消炎活性。
Objective To determine the chemical constituents of Lonicerae japonicae flos and their antibacterial and anti-inflammation activity.Methods The chemical components of Lonicerae japonicae flos were separated and purified by chromatography,and the structures of those compounds were further identified by spectroscopy.The anti-inflammation activity was evaluated by suppression against expression of nitric oxide(NO)and cyclooxygenase.At the same time,microdilution method was used for in vitro evaluation of antibacterial activities of conventional bacteria and anaerobic bacteria.Results Totally 22 compounds were identified as secoxyloganin(1),secologanol(2),kingiside(3),8-epikingiside(4),sweroside(5),ketologanin(6),vogeloside(7),epi-vogeloside(8),threoninosecologanin(9),5S-carboxystrictosidine(10),(2E,6S)-8-[α-L-arabinopyranosyl-(1’’→6’)-β-D-glucopyranosyl]-2,6-dimethyloct-2-eno-1,2’’-lactone(11),1,2-O-didicaffeoyl-cychopenta-3-ol(12),4,5-O-dicaffeoylquinic acid(13),caffeic acid(14),chlorogenic acid(15),syringin(16),rutin(17),supinaionodie B(18),cordysinin B(19),guanosine(20),adenosine(21),and nicotiamide(22).The isolated compounds had no evident inhibition on NO production and cyclooxygenase 1 and 2.Compounds 2,5,11,and 19 obviously inhibited anaerobic bacteria Streptococcus mutans,and compounds 4,6,8,and 15 showed certain inhibition on Staphylococcus aureus.Conclusion Twenty-two compounds have been identified from Lonicerae japonicae flos,and compounds 11,12,18 and 22 have been obtained for the first time.Compounds 2,4,5,6,8,11,15,and 19 from Lonicerae japonicae flos have certain bacteriostatic effect.
