复方鹿茸超微混悬酒工艺优化及对血虚小鼠模型的影响研究

摘要 目的优化复方鹿茸超微混悬酒制备工艺,并对其补血作用进行研究。方法将鹿茸超微粉碎配制为鹿茸药液;采用单因素考察法结合正交试验法优化提取条件,配制不含鹿茸的药液;二者混合得到复方鹿茸超微混悬酒。采用乙酰苯肼与环磷酰胺联合应用方法制备血虚模型,观察各组小鼠体重变化、游泳时间、外周血常规、免疫器官指数的差异,探讨不同给药剂量对小鼠补血的影响。结果配制的鹿茸药液为类乳白色混悬液。提取条件为:乙醇浓度60%、液料比20倍量、提取时间1 h、提取次数3次;配制不含鹿茸的药液为深红棕色药液。补血试验结果表明:与模型组比较,中剂量组与高剂量组可增加小鼠体重(P<0.01),延长游泳时间(P<0.01或P<0.05),升高外周血象中红细胞、血红蛋白、红细胞压积、血小板指数(P<0.01或P<0.05),降低白细胞、单核细胞百分比(P<0.01或P<0.05),提高免疫器官中胸腺重量与胸腺指数(P<0.05),降低脾脏重量与脾脏指数(P<0.01或P<0.05),降低肝脏重量与肝脏指数(P<0.01或P<0.05)。结论复方鹿茸超微混悬酒制备工艺设计合理,简便可行;能明显改善小鼠血虚症候,具有确切补血功效。 Objective To optimize the preparation technology of Compound Deer Antler Ultra-fine Suspension Liquor and to study its blood tonic effect.Methods The deer antler liquid was prepared by ultrafine crushing.Single factor investigation and orthogonal experiment were used to optimize the extraction conditions,and the medicine solution without pilose antler was prepared.Compound Deer Antler Ultra-fine Suspension Liquor was obtained by mixing them.The blood deficiency model was prepared by combined application of acetyl phenylhydrazine and cyclophosphamide.The changes in body weight,swimming time,peripheral blood routine and immune organ index of mice in each group were observed,and the effects of different doses on blood enrichment in mice were discussed.Results The prepared liquid of pilose antler was milky white suspension.The extraction conditions were as follows:60%ethanol,liquid-solid ratio of 20,1 h each time for extraction and total 3 times.The solution without deer antler was dark red brown solution.Results of blood enrichment test showed that:compared with model group,middle dose group and high dose group could increase body weight(P<0.01),prolong swimming time(P<0.01 or P<0.05),increase erythrocyte,hemoglobin,hematocrit and platelet index in peripheral blood imaging(P<0.01 or P<0.05).The percentages of WBC and monocyte decreased(P<0.01 or P<0.05),thymus weight and thymus index in immune organs increased(P<0.05),spleen weight and spleen index decreased(P<0.01 or P<0.05),and liver weight and liver index decreased(P<0.01 or P<0.05).Conclusion The preparation process of Compound Deer Antler Ultra-fine Suspension Liquor is reasonable,simple and feasible.It can obviously improve the syndrome of blood deficiency in mice and has the effect of enriching blood.
