Data Mining and Mechanism Analysis of Core Drug Use of Traditional Chinese Medicine in Children with Bronchial Asthma During Acute Attack

摘要 目的利用数据挖掘及网络药理学技术探究儿童支气管哮喘急性发作期中医核心用药及其作用机制。方法收集儿童支气管哮喘急性发作期中医治疗的医案,借助中医传承辅助平台获取核心用药。基于TCSMP数据库获取核心药物的主要活性成分,通过Swiss Target Prediction数据库和SEA数据库预测主要活性成分的靶点;在TTD数据库、Drugbank数据库检索支气管哮喘的相关靶点,将药物靶点与疾病靶点映射,获得核心药物治疗支气管哮喘的靶点,导入STRING数据库构建PPI网络并通过Cytoscape 3.2.1软件进行拓扑参数分析。通过DAVID数据库对核心靶点进行GO分析。结果查阅相关书籍25本,纳入医案130篇,获取4味核心药物及其主要化学成分150个,预测药物靶点数目为601个。获取支气管哮喘相关疾病靶点172个,核心药物治疗支气管哮喘的靶点79个。GO分析获得273条通路。结论儿童支气管哮喘急性发作期中医治疗核心用药为麻黄、苦杏仁、紫苏子和甘草,其作用机制可能与炎症应答、G蛋白偶联受体信号通路、类固醇激素生物合成等多通路相关。 Objective To explore the core drug use of traditional Chinese medicine in children with bronchial asthma during acute attack by using data mining and network pharmacology methods.Methods To collect children bronchial asthma acute phase of traditional Chinese medicine treatment of basis,with the aid of inheritance of traditional Chinese medicine auxiliary core platform for drug use.Based on TCSMP database access to the core of the main active ingredients,by Swiss Target Prediction database and SEA database to predict the main active ingredient of targets;Bronchial asthma in providing database,Drugbank database retrieval relevant targets,targets of forecast drug targets and disease mapping,get the core target of bronchial asthma drug treatment,import the STRING database construction of PPI network and the topology with the Cytoscape3.2.1 software parameter analysis.GO through DAVID database to analysis core targets.Results 25 related books,check into 130 basis,obtain four core drugs and its main chemical components of 150,to predict the number of drug targets for 601.Targets for bronchial asthma related diseases of 172,core drug target of 79 for the treatment of bronchial asthma.GO analysis obstained 273 pathway.Conclusion Children bronchial asthma acute phase of traditional Chinese medicine treatment of core drugs as Mahuang(Ephedrae Herba),Kuxingren(Armeniacae Semen Amarum),Zisuzi(Perillae Fructus)and Gancao(Glycyrrhizae Radix Et Rhizoma),its mechanism may be related to the inflammatory response,G protein coupled receptor signaling pathways,steroid hormone biosynthesis pathway.
