摘要
目的 探讨甲胎蛋白(AFP)、丝/苏氨酸蛋白激酶(AKT)2、核基质结合区结合蛋白(SATB)1在原发性肝癌患者中的表达水平及与临床病理特征的关系。方法 收集2019年5月至2020年5月于唐山市人民医院接受手术治疗的82例原发性肝癌患者的病历资料进行回顾性研究,测定癌组织及癌旁组织AFP mRNA、AKT2 mRNA、SATB1 mRNA表达水平,分析癌组织AFP mRNA、AKT2 mRNA、SATB1 mRNA表达水平与临床病理特征的关系,对比术后1年复发与未复发患者癌组织AFP mRNA、AKT2 mRNA、SATB1 mRNA表达水平,采用Logistic回归分析原发性肝癌患者术后复发的影响因素,采用受试者工作特征(ROC)曲线分析癌组织AFP mRNA、AKT2 mRNA、SATB1 mRNA对术后复发的预测价值。结果 癌组织AFP mRNA、AKT2 mRNA、SATB1 mRNA表达水平高于癌旁组织(P<0.05);癌组织AFP mRNA、AKT2 mRNA、SATB1 mRNA表达水平与TNM分期、病理分级有关(P<0.05);复发者癌组织AFP mRNA、AKT2 mRNA、SATB1 mRNA表达水平高于未复发者(P<0.05);癌组织AFP mRNA、AKT2 mRNA、SATB1 mRNA表达水平升高均为术后复发的影响因素(P<0.05);癌组织AFP mRNA、AKT2 mRNA、SATB1 mRNA联合预测复发的曲线下面积为0.853,灵敏度为89.47%,特异度为71.43%。结论 原发性肝癌患者癌组织AFP mRNA、AKT2 mRNA、SATB1 mRNA表达水平与TNM分期、病理分级及术后复发有关,三者联合检测可为临床预测术后复发提供有效的量化参考依据,亦可能为原发性肝癌治疗提供新的靶点。
Objective To explore the expression levels of alpha-fetoprotein(AFP),serine/threonine protein kinase 2(AKT2) and nuclear matrix binding domain binding protein 1(SATB1) in the patients with primary liver cancer and their relationship with clinicopathological characteristics.Methods The case data of 82 patients with primary liver cancer receiving surgery in the Tangshan Municipal People′s Hospital from May 2019 to May 2020 were selected for conducting the retrospective study.The expression levels of AFP mRNA,AKT2 mRNA and SATB1 mRNA in cancer tissues and paracancerous tissues were measured.The relationship between the expression levels of AFP mRNA,AKT2 mRNA and SATB1 mRNA in cancer tissues with the clinicopathological characteristics were analyzed.The expression levels of AFP mRNA,AKT2 mRNA and SATB1 mRNA in cancer tissues were compared between the patients with recurrence and the patients without recurrence in postoperative 1 year.The Logistic regression was used to analyze the influencing factors of postoperative recurrence in patients with primary liver cancer, and the predictive values of AFP mRNA,AKT2 mRNA,SATB1 mRNA in cancer tissue for postoperative recurrence were analyzed by adopting the receiver operating characteristic(ROC) curve.Results The expression levels of AFP mRNA,AKT2 mRNA and SATB1 mRNA in cancer tissues were higher than those in paracancerous tissues(P<0.05);the expression levels of AFP mRNA,AKT2 mRNA and SATB1 mRNA in cancer tissues were related to the TNM staging and pathological grading(P<0.05);the expression levels of AFP mRNA,AKT2 mRNA and SATB1 mRNA in cancer tissues of the patients with recurrence were higher than those of the patients without recurrence(P<0.05);the high expression levels of AFP mRNA,AKT2 mRNA and SATB1 mRNA in cancer tissues were all influential factors for postoperative recurrence(P<0.05);the area under the curve of the combination detection of AFP mRNA,AKT2 mRNA and SATB1 mRNA in cancer tissues for predicting recurrence was 0.853,the sensitivity was 89.47% and the s
