摘要
为了研究感染猪伪狂犬病病毒(Pseudorabies virus,PRV)后,脾脏中T/B淋巴细胞亚群的变化情况,试验将40只Balb/c小鼠均分为对照组和试验组,试验组分为低、中、高剂量组,分别于皮下接种1×10^(3)TCID_(50)/100μL、1×10^(4)TCID_(50)/100μL和1×10^(5) TCID_(50)/100μL PRV-HLJ毒液200μL,对照组注射等量PBS,观察动物生存情况,确定最佳接种剂量。在此基础上,选取80只Balb/c小鼠,分成对照组和试验组,分别于感染PRV-HLT后24,48,72,96小时采集各组小鼠脾脏,制备细胞悬液,采用流式细胞术检测T/B淋巴细胞亚群的变化。结果表明:在感染PRV-HLT后96小时高、中剂量组小鼠全部死亡,低剂量组小鼠存活率为50%,确定最佳接种剂量为1×10^(3)TCID_(50)/100μL。与对照组比较,感染PRV-HLJ后24,48,72小时,小鼠脾脏中CD3^(+)CD4^(+)T淋巴细胞比例极显著下降(P<0.01),并于感染后96小时开始回升,但仍差异极显著(P<0.01);CD3^(+)CD8^(+)T淋巴细胞和CD19^(+)B淋巴细胞比例在整个感染期间极显著下降(P<0.01),变化规律与CD3^(+)CD4^(+)T淋巴细胞基本一致;CD4^(+)/CD8^(+)比值增加,但各组间差异不显著(P>0.05)。说明PRV感染小鼠后,其脾脏中T/B淋巴细胞亚群及比例下调,对外周免疫功能比例有一定的抑制作用。
In order to study the changes of T/B lymphocyte subsets in the spleen after infection with porcine Pseudorabies virus(PRV),in the experiment,40 Balb/c mice were equally divided into control group and experimental groups.The experimental groups were divided into low dose,medium dose and high dose groups,which were inoculated subcutaneously with the titers of 1×10^(3)TCID_(50)/100μL,1×10^(4)TCID_(50)/100μL and 1×105 TCID_(50)/100μL respectively in PRV-HLJ virus solution 200μL;the control group was injected with the same volume of PBS.The survival of the animals was observed to determine the optimal inoculation dose.Based on the above results,80 Balb/c mice were selected and divided into control group and experimental groups.The spleens of mice in each group were collected 24,48,72,and 96 h after PRV-HLT infection,respectively,to prepare cell suspensions,and flow cytometry was used to detect the changes of T/B lymphocyte subsets.The results showed that all the mice in the high and medium dose groups died 96 h after infection with PRV-HLT,and the survival rate of the mice in the low dose group was 50%.The optimal inoculation dose was determined to be 1×10^(3)TCID50/100μL.Compared with the control group,the proportion of CD3^(+)CD4^(+)T lymphocytes in the spleen of mice decreased significantly 24,48,and 72 h after infection with PRV-HLJ(P<0.01),and began to rise 96 h after infection with very significant difference(P<0.01).The ratio of CD3^(+)CD8^(+)T lymphocytes to CD19^(+)B lymphocytes decreased significantly during the whole infection period(P<0.01),and the change rule was consistent with the CD3^(+)CD4^(+)T lymphocyte gene.The CD4^(+)/CD8^(+)ratio increased,but the difference was not significant(P>0.05).The results indicated that after PRV infection in mice,the T/B lymphocyte subsets and proportions in the spleen were down-regulated,which had a certain inhibitory effect on peripheral immune function.
