摘要
全蝽(Walker,1868)隶属于半翅目蝽科,主要为害苹果及其他蔷薇科的果树。为丰富蝽科线粒体基因组学的基本数据,进一步阐明蝽科在蝽总科中的系统发育关系,确定全蝽在蝽科中的分类地位,通过Illumina Miseq平台测序、组装拼接后进行全蝽线粒体基因的注释和分析,结合NCBI数据库中已有数据,基于贝叶斯法(BI)和最大似然法(ML)构建系统发育树。结果表明,全蝽完整的线粒体基因长度为15479 bp,其中包含37个基因(13个蛋白编码基因、22个tRNA基因和2个rRNA基因)和一个控制区;全蝽的线粒体基因组的基因顺序与典型的蝽科昆虫一致。经分析,全蝽的A+T总含量为76.18%,远大于G+C含量,表现为明显的AT偏斜;大多数蛋白质编码基因的起始密码子为ATN和TTG,终止密码子为TAN或单个T碱基。除trnA和trnS1外,其他20个tRNA基因的二级结构都能折叠成一个典型的三叶草形状。蛋白编码基因进化速率表明,atp8进化速率最大,进化最快,cox1进化最慢。系统发育树结果显示,全蝽位于蝽科支系且蝽科为单系,这与先前基于形态学或部分分子片段所构建的蝽总科系统发育树结果一致。
Homalogonia obtusa(Walker,1868)belongs to Hemiptera Pentatomidae,mainly damages apple and other species of Rosaceae.To enrich the basic data of mitochondrial genomics of Pentatomidae,clarify the phylogenetic relationship of Pentatomidae family in Pentatomoidea,and define the taxa of H.obtusa in Pentatomidae,in this study,the complete mitochondrial genome of Homalogonia obtusa was annotated and analyzed by using Illumina Miseq platform and assembly and splicing.The phylogenetic tree was constructed based on Bayesian Inference(BI)and Maximum Likelihood method(ML),using the existed data from NCBI database.The results showed that the length of the complete mitochondrial gene of Homalogonia obtusa was 15479 bp,of which 37 typical mitochondrial genes(13 protein-coding genes,22 tRNA genes,and 2 rRNA genes)and a control region were contained.The gene order of the mitogenome of H.obtusa was consistent with typical pentatomid insects.By analysis,the overall content of A+T of H.obtusa was 76.18%,much greater than the content of G+C,showing obvious AT deviation.Most protein-coding genes started with ATN and TTG,and stopped at TAN or a single T.Other 20 tRNA genes folded into a typical cloverleaf secondary structure except for trnA and trnS1.The evolution rate analyses of protein coding genes showed that the evolutionary rate of atp8 was the fastest,and cox1 was the slowest.The phylogenetic tree results showed H.obtusa belonged to the branch of Pentatomidae,and Pentatomidae was monophyletic,which was coincident with the result of previous phylogenetic tree of Pentatomoidea constructed based on morphological characteristics and partial molecular fragments.
