摘要
ERF基因家族在乙烯信号转导和内源乙烯合成的过程中扮演着重要角色。为探究ERF在外源乙烯处理过程中的响应及表达调控机制,本实验对芒果转录组序列进行测序分析,从获得的Unigenes中筛选出31个具有AP2保守结构域的ERF基因家族成员,并对这些基因进行了序列分析,预测了这些基因所含有的motif结构。结果表明,这些基因都属于亲水蛋白,但在其他理化性质方面存在着较大差别;在多序列比对后,发现该家族成员都含有YRG元件和RAYD元件。6个芒果ERF家族基因的表达情况显示,MiERF-AP2-5、MiERF-AP2-6、MiERF-AP2-17、MiERF-AP2-20及MiERF-AP2-29在外源乙烯处理后的表达量均有所上升,预示着这5个基因在外源乙烯处理条件下可能对其代谢起到正向调控的作用,而MiERF-AP2-9在处理后却出现下降趋势,预示着该基因或许是起负向调控作用或是扮演着转录抑制因子的角色。
The ERF gene family plays an important role in ethylene signal transduction and endogenous ethylene synthesis.In order to investigate the response of ERFs to exogenous ethylene treatment and the mechanism of expression regulation,we sequenced the mango transcriptome sequences and screened 31 ERF gene family members with conserved structural domains of AP2 from the obtained Unigenes,analyzed their physicochemical properties,further analyzed the affinities of these genes,and predicted the motifs contained in these genes structure.The results showed that all these genes belonged to hydrophilic proteins,but differed significantly in other physicochemical properties;after multiple sequence alignment,the family members were found to contain YRG elements and RAYD elements.The expression of MiERF-AP2-5,MiERF-AP2-6,MiERF-AP2-17,MiERF-AP2-20 and MiERFAP2-29 increased after exogenous ethylene treatment,suggesting that these five genes may play a positive role in regulating their metabolism under exogenous ethylene treatment,while MiERF-AP2-9 showed a decreasing trend after treatment,suggesting that this gene may play a negative regulatory role or act as a transcriptional repressor.
