摘要
目的:探讨硼酸钠对人肝癌Bel-7402细胞基因表达的影响。方法:用4 mmol/L硼酸钠处理Bel-7402细胞2 h和24 h。提取细胞总RNA,使用基因芯片进行基因表达分析,然后进行基因GO分析和Kegg信号通路分析。结果:与对照组相比,硼酸钠作用2 h和24 h后,Bel-7402细胞多个基因发生改变。2 h组有462个差异表达基因(270个上调、192个下调);24 h组有1917个差异表达基因(980个上调、937个下调)。GO分析显示硼酸钠引起Bel-7402细胞的差异基因,涉及到分子功能、生物学过程多个层级。Kegg通路富集分析显示,硼酸钠作用2 h后差异表达基因参与多条信号通路(NF-κB、MAPK等),作用24 h后差异表达基因主要参与NF-κB信号通路、细胞凋亡等。结论:硼酸钠处理Bel-7402细胞后,导致多种基因表达上调或下调,从而发挥多种生物学作用。
Obiective To study the effect of borax on gene expression of human hepatoma Bel-7402 cells. Methods Bel-7402 cells were treated with 4 mmol/L borax for either 2 or 24 h. Total RNA was extracted, and gene expression analysis was performed using GeneChip, followed by gene GO analysis and Kegg signaling pathway analysis. Results Compared with the control group, many genes were changed in Bel-7402 cells after borax treatment for 2 h and 24 h. There were 462 differentially expressed genes(270 up-regulated and 192 down regulated) in 2 h group and there were 1917 differentially expressed genes(980 up-regulated and 937 down regulated) in the 24 h group. GO analysis showed that borax caused differential genes in BEL-7402 cells, involving multiple levels of molecular functions and biological processes. Kegg pathway enrichment analysis showed that the differentially expressed genes were involved in multiple signaling pathways(NF-κB, MAPK, etc.) after 2 h of sodium borate exposure, and the differentially expressed genes were mainly involved in NF-κB signaling pathway and cell apoptosis after 24 h of exposure. Conclusion Bel-7402 cells treated with sodium borate can up-regulate or down-regulate the expression of various genes, thereby exerting various biological effects.
