光调控对粗肋草试管苗愈伤组织诱导的影响 被引量:5

Effect of Light Regulation on Callus Induction System of Test-tube Plantlets of Aglaonema commutatum Schott

摘要 以‘如意皇后’和‘烟花’粗肋草生长健壮、无污染的试管苗为试验试材,选取带侧芽的茎段为外植体,通过调控光环境,研究光周期、光照强度、照光方式以及光质(采用LED灯管为光源)配比对粗肋草愈伤组织诱导及出愈率的影响。结果表明:诱导8周后,‘如意皇后’光周期为12 h/d、‘烟花’光周期为24 h/d时,愈伤组织诱导率最高。光强为3000 lx时‘如意皇后’出愈率最高,光强为2500 lx时‘烟花’出愈率最高。暗培养30 d后光培养30 d的照光方式下,粗肋草的出愈率最高。而为30%蓝光+70%红光(B∶R=3∶7)条件下,‘如意皇后’和‘烟花’的出愈率最高生长状况最佳,光质配比为100%红光(100%R)、100%蓝光(100%B)时,诱导愈伤褐化严重,愈伤诱导率低。本研究初步筛选出适宜粗肋草愈伤诱导的光环境条件,可进一步优化改良,为今后粗肋草试管苗愈伤诱导提供技术参考。 Taking the growing robustly and pollution-freetest-tube seedlings of’Red Valentine’and’Firework’as test materials.Taking the rhizome with axillary buds as the explants,by regulating the light environment,to study the effects of photoperiods,illumination intensity,light methods,and light quality ratios(LED tube as light source)on callus induction and recovery rate of Aglaonema commutatum Schott.Experimental results show that:8 weeks after induction,the photoperiod of’Red Valentine’is 12 h/d and the photoperiod of’Firework’is 24 h/d,the callus induction rate is the highest.The illumination intensity is 3000 lx,the’Red Valentine’has the highest recovery rate,and the illumination intensity is 2500 lx,the’Firework’has the highest recovery rate.After dark culture for 30 days,light culture for 30 days,the callus induction rate is the highest of Aglaonema commutatum Schott.The ratio of light quality was 30%blue light+70%red light(B∶R=3∶7),’Red Valentine’and’Firework’have the highest healing rate and the best growth conditions,the ratio of light quality is 100%red light(100%R)and 100%blue light(100%B),the induced callus browning was serious,low callus induction rate.This study has initially screened out suitable light environment conditions for callus induction,it can be further optimized and improved to provide technical reference for callus induction of test-tube plantlet.
