摘要
本研究旨在分析浙江红花油茶种质材料的遗传多样性,挖掘与重要经济性状密切关联的SSR分子标记。以6个产地共45份经济性状变异较大的浙江红花油茶种质材料为研究对象,开展遗传多样性评估与群体遗传结构分析,并基于TASSEL软件中的广义线性模型(GLM)与混合线性模型(MLM)相结合的方法,利用49个多态性较高的EST-SSR标记与17个经济性状进行关联分析。研究结果显示,49对SSR引物共检测到199个等位基因,标记的PIC多态性信息指数平均值达到0.546 7,受检测群体的基因多样度平均值为0.605 2,显示较高水平的多样性;群体遗传结构分析将浙江红花油茶种质分为3个亚群,表明群体遗传背景单一。关联分析中,GLM法共检测到21个SSR标记与浙江红花油茶的果皮厚、千粒籽重、鲜出籽率、油酸、硬脂酸等9个性状极显著关联(P<0.01),表型变异解释率在13.51%~56.55%之间;而在这些标记中,MLM方法则可检测到11个SSR标记与果皮厚、鲜出籽率、亚麻酸、油酸、硬脂酸、总脂肪酸等9个性状显著关联(P<0.05),表型变异的贡献率为20.39%~57.36%,其中与油品质相关的标记有6对,分别是CC_e SSR03、CC_eSSR29、CC_eSSR77、CC_eSSR78、CC_eSSR85、CC_eSSR327。研究可为浙江红花油茶重要经济性状的基因定位和分子标记辅助育种提供技术基础。
The purpose of this study was to analyze the genetic diversity of C. chekiangoleosa germplasm materials,and to discover SSR molecular markers closely related to important economic traits. A total of 45 C. chekiangoleosa germplasm materials with large variations in economic traits from 6 producing areas were used as the object of study to carry out genetic diversity assessment and population genetic structure analysis. 49 highly polymorphic EST-SSR markers and 17 economic traits were used for association analysis through the two methods of combining general linear model(GLM) and mixed linear model(MLM) in tassel software. The results showed that a total of 199 alleles were detected by 49 SSR primers. The average PIC polymorphism information index of the markers was 0.546 7, and the average gene diversity of the tested population was 0.605 2, indicating a high level of diversity. The population structure analysis suggested that the germplasm of C. chekiangoleosa was divided into 3subgroups, showing that the population genetic background was single. The association analysis indicated that the GLM method detected 21 SSR markers with extremely significant correlations with 9 traits such as fruit skin thickness, TKW, fresh seed yield, oleic acid, and stearic acid of C. chekiangoleosa(P<0.01), which explained the phenotypic variation with a range of 13.51% to 56.55%;the MLM method showed that 11 SSR markers were significantly associated with 9 traits such as fruit skin thickness, fresh seed yield, linolenic acid, oleic acid, stearic acid and total fatty acid among these markers(P<0.05), which explained the phenotypic variation with a range of20.39% to 57.36%;Six pairs of markers related to oil quality were CC_eSSR03, CC_eSSR29, CC_eSSR77,CC_eSSR78, CC_eSSR85, CC_eSSR327. The study can lay the technical foundation for the gene mapping and molecular marker-assisted breeding of the important economic traits of C. chekiangoleosa.
