摘要
目的 分析妊娠期高危型人乳头瘤病毒(hr-HPV)感染患者胎盘组织中转化生长因子β(TGFβ)-Smad信号通路表达水平与子痫发生的关系。方法 选取2017年12月-2020年12月在莱西市人民医院行规律产检和分娩的314例妊娠期hr-HPV感染患者为HPV感染组,同期在医院进行产检和分娩的314名健康孕妇为对照组。收集两组一般资料、子痫和子痫前期发生情况,采用酶联免疫吸附试验法检测血清肿瘤坏死因子α(TNF-α)、白细胞介素(IL)-6和IL-10水平,蛋白印迹法(Western blot)检测胎盘组织TGFβ-Smad信号通路蛋白表达。结果 HPV感染组患者血清TNF-α、IL-6和L-10水平高于对照组(P<0.05),IL-2水平低于对照组(P<0.05),胎盘组织TGF-β1、p-Smad2/Smad2和p-Smad3/Smad3蛋白表达高于对照组(P<0.05),Smad7蛋白表达低于对照组(P<0.05);HPV感染组患者子痫前期和子痫发生率高于对照组(P<0.05);HPV感染组患者中子痫组胎盘组织TGF-β1、p-Smad2/Smad2和p-Smad3/Smad3蛋白表达高于非子痫组(P<0.05),Smad7蛋白表达低于非子痫组(P<0.05),血清TNF-α、IL-6和IL-2水平高于非子痫组(P<0.05),IL-10水平低于非子痫组(P<0.05)。结论 妊娠期hr-HPV感染患者胎盘组织TGFβ-Smad信号通路表达和免疫炎症因子水平升高,且其升高水平与子痫和子痫前期发生有关。
OBJECTIVE To observe the relationship between the expression of transforming growth factor β-Smad(TGFβ-Smad) signaling pathway in placenta tissues and the occurrence of eclampsia in patients with high-risk human papilloma virus(hr-HPV) infection during pregnancy. METHODS A total of 314 patients with hr-HPV infection during pregnancy who underwent regular antenatal examination and gave birth in Laixi People’s Hospital from Dec 2017 to Dec 2020 were assigned as the infection group, meanwhile, 314 healthy pregnant women who underwent regular antenatal examination and gave birth were set as the control group. The baseline data and incidence of eclampsia and preeclampsia were collected from the two groups. The levels of serum tumor necrosis factorα(TNF-α), interleukin(IL)-6 and IL-10 were detected by enzyme-linked immunosorbent assay, and the expression of TGFβ-Smad signaling pathway protein in placenta tissues was detected by Western blot. RESULTS The levels of serum TNF-α, IL-6 and L-10 of the HPV infection group were significantly higher than those of the control group(P<0.05), the IL-2 level of the HPV infection group was significantly lower than that of the control group(P<0.05). The expression levels of TGF-β1, p-Smad2/Smad2 and p-Smad3/Smad3 in placenta tissues were significantly higher in the HPV infection group than in the control group(P<0.05),the expression level of Smad7 protein of the HPV infection group was significantly lower than that of the control group(P<0.05). The incidence rates of preeclampsia and eclampsia of the HPV infection group were significantly higher than those of the control group(P<0.05). Among the patients with HPV infection, the expression levels of TGF-β1, p-Smad2/Smad2 and p-Smad3/Smad3 proteins in placenta tissues of the eclampsia group were significantly higher than those of the non-eclampsia group(P<0.05), the expression level of Smad7 protein of the eclampsia group was lower than that of the non-eclampsia group(P<0.05);the levels of serum TNF-α, IL-6 and IL-2 of the
