响应面法优化黄晶果果皮总黄酮提取工艺及抗氧化活性 被引量:10

Optimization of Extraction Process by Response Surface Methodology and Antioxidant Activity of Total Flavonoids from Abiu.Peels

摘要 文章研究黄晶果果皮总黄酮提取工艺,并评价其抗氧化活性。采用超声波辅助法提取黄晶果果皮总黄酮,通过单因素、响应面法优化工艺,通过总黄酮的还原能力、DPPH法、Fenton反应法、邻苯三酚自氧化法和ABTS法评价其抗氧化活性。结果表明:最优工艺参数为为液料比30 mL/g、乙醇浓度79%、提取时间20 min、提取温度52℃,此条件下黄晶果果皮总黄酮提取率达1.70%。当总黄酮提取液浓度为0.3 mg/mL时,其吸光度为0.954,这表明总黄酮提取液具备一定的铁离子还原能力;对DPPH、羟基、超氧阴离子和ABTS自由基的半数清除率IC_(50)值分别为0.67、1.56、0.03、0.17 mg/mL。这表明黄晶果果皮总黄酮具有较强的抗氧化活性,可为其进一步开发成天然抗氧化剂提供理论依据。 In order to study the extraction process of total flavonoids from Abiu.peels and evaluate its antioxidant activity,ultrasonic-assisted extraction was used to extract total flavonoids from these peels.The extraction process of total flavonoids from Abiu.peels was optimized by single factor and response surface methodology.The antioxidant activities of Abiu.peels total flavonoids were tested by the reducing ability of total flavonoids,DPPH assay,Fenton’s oxidation reaction,pyrogallol autoxidation method and ABTS assay.The results showed that the optimum technological parameters were as follows:liquid-material ratio 30 mL/g,ethanol concentration 79%,extraction time 20 min,extraction temperature 52℃.Under these conditions,the extraction rate of total flavonoids from Abiu.peels was 1.70%.When the concentration of total flavonoids extract was 0.3 mg/mL,its absorbance was 0.954,which indicated that the total flavonoids in the Abiu.peels had a certain reducing ability of iron ion;The IC_(50) values of the total flavonoids in the Abiu.peels to DPPH free radical,hydroxyl free radical,superoxide anion free radical and ABTS free radical were 0.67 mg/mL,1.56 mg/mL,0.03 mg/mL and 0.17 mg/mL scavenging rates respectively.The results showed that the total flavonoids in the Abiu.peels had strong antioxidant activity,which could provide a theoretical basis for its further development of natural antioxidants.
