摘要
以桑黄子实体为原料,经蒸汽爆破预处理,并采用超声辅助提取法,以总多糖和总多酚的得率为考察指标,确定蒸汽爆破处理最适条件;以DPPH和ABTS自由基清除率以及胆酸盐吸附率为考察指标,进行活性评价。结果表明:桑黄多糖最适蒸汽爆破条件为1.50 MPa、180 s;桑黄多酚最适蒸汽爆破条件为1.50 MPa、150 s;经最适蒸汽爆破条件处理后,桑黄多糖和多酚提取物DPPH自由基清除率分别为(50.14±4.43)%和(82.78±5.12)%,ABTS自由基清除率分别为(34.82±1.73)%和(64.22±5.34)%;未经蒸汽爆破处理的多糖提取物和蒸汽爆破处理的多酚提取物均显示一定的胆酸盐吸附能力,吸附率分别为(17.81±0.18)%和(9.06±0.02)%;协同(蒸汽爆破多糖:蒸汽爆破多酚为1:3)显示较强的清除DPPH自由基活性,清除率可达(67.80±9.45)%;协同(蒸汽爆破多糖:蒸汽爆破多酚为1:4)显示较强的清除ABTS自由基活性,清除率可达(56.98±6.23)%;协同(未蒸汽爆破多糖:蒸汽爆破多酚为1:1)显示一定的胆酸盐吸附能力,吸附率可达(19.14±0.97)%;蒸汽爆破预处理促进了桑黄多糖、多酚的释放并提高了其抗氧化活性,结果可为桑黄子实体深度开发利用提供参考依据。
Taking the fruiting bodies of Inonotus hispidus as raw materials,steam explosion(SE)pretreatment,ultrasonic-assisted extraction method and the yield of total polysaccharides and total polyphenols as the index,the optimum conditions of steam explosion treatment were determined.DPPH radical scavenging,ABTS radical scavenging and bile salt adsorption were used to evaluate the activity of the functional components.The results showed that the optimal processing condition of steam explosion for polysaccharide extract from Inonotus hispidus was 1.50 MPa and 180 s.The optimal processing condition of steam explosion for polyphenols extract from Inonotus hispidus was 1.50 MPa and 150 s.Under treatment with the optimal steam explosion conditions,the DPPH radical scavenging activity of polysaccharides and polyphenols extracts from Inonotus hispidus were(50.14±4.43)%and(82.78±5.12)%,respectively.The ABTS radical scavenging activity of polysaccharides and polyphenols extracts from Inonotus hispidus were(34.82±1.73)%and(64.22±5.34)%,respectively;polysaccharides without steam explosion and polyphenols with steam explosion extract showed certain activity of bile salt adsorption,were(17.81±0.18)%and(9.06±0.02)%,respectively;coordinated sample(polysaccharides with SE:polyphenols with SE was 1:3)showed strong activity of DPPH radical scavenging,up to(67.80±9.45)%Coordinated sample(polysaccharides with SE:polyphenols with SE was 1:4)showed strong activity of ABTS radical scavenging,up to(56.98±6.23)%;coordinated sample(polysaccharides without SE:polyphenols with SE was 1:1)showed certain activity of bile salt adsorption,up to(19.14±0.97)%.Steam explosion pretreatment promoted the release of polysaccharides and polyphenols from Inonotus hispidus,and enhanced the antioxidant activity.This study provides a reference for the further development and utilization of fruitbody of Inonotus hispidus.
