朴实方对功能性便秘小鼠排便、结肠动力及结肠组织学特征的影响 被引量:1

Effects of Pushi Decoction on Defecation, Colonic Motility, and Molonic Histological Characteristics in Mice with Functional Constipation

摘要 目的 探讨朴实方对功能性便秘小鼠模型排便及结肠动力的影响。方法 将50只SPF级雄性ICR小鼠随机分为正常组,模型组,朴实方高、低剂量组,阳性对照组,每组各10只。除正常组外,其余4组小鼠均以复方地芬诺酯混悬液10mg/(kg·d)连续灌胃2周建立小鼠功能性便秘模型。造模成功后,朴实方高、低剂量组分别按照18.2、9.1 g/(kg·d)的中药剂量进行灌胃,阳性对照组以普卢卡必利混悬液(每天0.3 mg/kg)灌胃,正常组、模型组则以等体积生理盐水灌胃,疗程2周。观察、记录各组小鼠排便次数、大便形态变化、肠道碳推进率等,采用免疫组化法观察、测量各组小鼠结肠黏液细胞数量及肌层厚度等指标变化。结果 与正常组比较,模型组小鼠的粪便粒数和含水率明显减少(P<0.01),肠道推进率降低(P<0.01),结肠黏液细胞数量显著减少(P<0.01),结肠肌层厚度降低(P<0.01);与模型组比较,中药两组及阳性对照组小鼠粪便粒数和含水率、肠道推进率均升高(P<0.01),结肠组织学特征也趋于正常;中药两组组间比较,小鼠的粪便粒数、含水率及肠道推进率差异无统计学意义(P>0.05);与阳性对照组比较,中药两组小鼠各项指标差异无统计学意义(P>0.05)。结论 朴实方通过改善结肠结构、促进结肠运动达到改善排便的作用。 Objective To observe the effect of Pushi Decoction(PSD)on defecation and colonic motility(FC) in mice with functional constipation(FC). Methods Totally 50 SPF grade male ICR FC mice were randomly divided into normal control group, FC model group, low and high dose PSD groups, and positive control group, 10 in each group. FC model was established by intragastric administration of Compound Diphenoxylate 10 mg/(kg · d) for 2 successive weeks in mice of the rest 4 groups(except the normal group). After successful modeling,PSD at 18.2 and 9.1 g/(kg · d) was administered to mice in high and low dose PSD groups by gastrogavage. Prucalopride suspension(0.3 mg/kg)was administered to mice in the positive control group by gastrogavage. Equal volume of normal saline was administered to mice in the normal group and the model group. All medication lasted for 2 weeks. The changes of defecation times, stool morphology, intestinal carbon propulsion rate and colon tissue were observed and recorded. Numbers of mucous cells in the colon and thickness of muscular layer were observed and determined by immunohistochemical assay.Results Compared with the normal control group, the fecal granule number and water content, and intestinal propulsion rate,numbers of mucous cells in the colon and thickness of muscular layer in the FC model group significantly decreased(P<0.01).Compared with the model group, the fecal granule number and water content, intestinal propulsion rate in the two PSD groups as well as positive control group significantly increased(P<0.01).The histological characteristics of colon tended to be normal. There were no statistical differences in the fecal granule number, water content,or intestinal propulsion rate between the two PSD groups(P>0.05). Compared with the positive control group, there were no statistical differences in any indicator of the two PSD groups(P>0.05). Conclusion PSD improved defecation by improving colon structure and promoting colon movement.
