摘要
目的分析程序性死亡受体-1(PD-1)/程序性死亡配体-1(PD-L1)、调节性T细胞(Treg)及其相关细胞因子在宫颈癌患者外周血和组织中的表达情况。方法选择2020年1月—2021年8月本院收治的19例宫颈癌患者,19例宫颈癌(CC)患者和23例宫颈上皮内瘤变(CIN)患者作为病例组,同期25名健康女性体检者为对照组(NC)。使用流式细胞法和酶联免疫吸附实验(ELISA)检测外周血Treg上PD-1、Treg相关细胞因子的表达情况。采用多重荧光免疫组化,检测宫颈癌组织中CD8^(+)PD-1、CD4^(+)Foxp3^(+)的表达,并以癌旁组织作为对照。结果宫颈癌组外周血CD4^(+)CD25^(+)CD127^(low/-)Treg、Treg上PD-1、PD-1、PD-L1、叉头翼状螺旋转录因子3(Foxp3)、白细胞介素-10(IL-10)、转化生长因子-β(TGF-β)水平均显著高于宫颈上皮内瘤变组和对照组(均P<0.05),宫颈癌组白细胞介素-2(IL-2)、干扰素-γ(IFN-γ)水平均低于宫颈上皮内瘤变组和对照组(均P<0.05)。宫颈癌组织中CD8^(+)PD-1、CD4^(+)Foxp3^(+)细胞数明显多于癌旁组织(均P<0.05)。结论宫颈癌患者外周血和组织中PD-1/PD-L1、Treg表达水平明显升高,外周血中存在Treg相关细胞因子的异常表达。PD-1/PD-L1、调节性T细胞可能在宫颈癌的发生、发展及免疫逃逸中发挥了重要作用。
Objective To analyze the expression of programmed death receptor-1(PD-1)/programmed death ligand-1(PD-L1),regulatory T cells(Treg)and related cytokines in peripheral blood and tissues of patients with cervical cancer.Methods 19 cases of patients suffered with cervical cancer those were admitted to our hospital during January 2020 and August 2021 were enrolled as study objects,and together with 23 cases of patients with cervical intraepithelial neoplasia(CIN)were selected as the case group,and 25 healthy women were selected as the control group(NC).The expression of PD-1 on Treg and Treg-related cytokines in peripheral blood were detected by flow cytometry and Enzyme linked immunosorbent assay(ELISA).The expression of CD8^(+)PD-1 and CD4^(+)Foxp3^(+) in cervical cancer tissues were detected by multiple fluorescent immunohistochemistry,and the para-cancer tissues were used as controls.Results The levels of CD4^(+)CD25^(+)CD127^(low/-)Treg,PD-1 on Treg,PD-1,PD-L1,fork-headed pterygoid transcription factor 3(Foxp3),IL-10,TGF-βin peripheral blood of cervical cancer group were significantly higher than those in CIN group and control group(all P<0.05).The levels of IL-2 and IFN-γin peripheral blood of cervical cancer group were significantly lower than those in CIN group and control group(all P<0.05).The numbers of CD8^(+)PD-1,CD4^(+)Foxp3^(+) cell in cervical cancer tissue were significantly higher than those in para-cancer tissue(all P<0.05).Conclusions The expression of PD-1/PD-L1 and Treg in peripheral blood and cancer tissues of patients with cervical cancer were significantly increased,and there was an abnormal expression of Treg-related cytokines in peripheral blood.PD-1/PD-L1 and regulatory T cells may play an important role in the occurrence,development and immune escape of cervical cancer.
