“强桑1号”的叶片、茎段愈伤组织诱导增殖研究

摘要 探究不同激素调节剂对“强桑1号”叶片和茎段愈伤组织诱导与增殖的影响,为建立高效、系统的组培快繁体系奠定良好的基础。以“强桑1号”的叶片、茎段为外植体进行愈伤组织的诱导增殖培养。实验结果表明:在单一激素6-苄氨基嘌呤(6-BA)、吲哚丁酸(IBA)和噻苯隆(TDZ)培养基配方中,2.41 mg/L WPM+1.62 mg/L或4.86 mg/L IBA+25 g/L蔗糖+8 g/L琼脂(pH=5.2)培养基中叶片和茎段的愈伤组织诱导效果最好,诱导率为100%;多激素培养基中,2.41 mg/L WPM+0.02 mg/L IBA+0.001 mg/L TDZ+0 mg/L 6-BA或0.075 mg/L 6-BA+25 g/L蔗糖+8 g/L琼脂(pH=5.2)中愈伤组织的诱导率为100%。在2.41 mg/L WPM+4.86 mg/L IBA+25 g/L蔗糖+8 g/L琼脂(pH=5.2)培养基中,叶片和茎段的愈伤组织生根效果最好,诱导率为97.22%。 Experiment was carried out to study the effects of different hormone regulators on callus induction and proliferation of"Qiangsang No.1"leaves and stems,so as to lay a good foundation for the establishment of efficient and systematic tissue culture and rapid propagation system.The leaf and stem segments of"Qiangsang No.1"were used as explants for callus induction and proliferation culture.The results showed that the callus induction effect of leaves and stems was the best in the medium with single hormone 6-Benzylaminopurine(6-BA)or indole butyric acid(IBA)or thiabenduron(TDZ)with 2.41 mg/l WPM+1.62 mg/l or 4.86 mg/l IBA+25 g/L sucrose+8g/L agar(pH=5.2),and the induction rate was 100%.The callus induction rate was also 100%in the multi-hormone medium with 2.41 mg/l WPM+0.02 mg/L IBA+0.001 mg/L TDZ+0 mg/L 6-BA or 0.075 mg/L 6-BA+25 g/L Sucrose+8g/L Agar(pH=5.2).The callus rooting effect of leaves and stems was the best,and the induction rate was 97.22%in the medium of 2.41 mg/l WPM+4.86 mg/l IBA+25 g/L Sucrose+8 g/L Agar(pH=5.2).
