防治东亚飞蝗的杀虫剂筛选及不同组织酶活分析

Screening of insecticides and analysis of detoxification enzyme activity in different tissues for control of Locusta migratoria manilensis(Meyen)

摘要 [目的]探究不同杀虫剂对东亚飞蝗Locusta migratoria manilensis(Meyen)的生物活性及对解毒酶活性的影响,为研究东亚飞蝗的抗药性管理提供依据。[方法]采用浸叶法测定6种杀虫剂对东亚飞蝗的生物活动,并测定LC浓度下东亚飞蝗不同组织解毒酶活性的差异。[结果]6种杀虫剂对东亚飞蝗的生物活性从高到低依次为溴氰虫酰胺、高效氯氟氰菊酯、啶虫脒、阿维菌素、吡虫啉及乙基多杀菌素。溴氰虫酰胺处理后,CarE活性在马氏管、中肠及脂肪体中显著提高,马氏管和中肠GST活性明显提高,马氏管和脂肪体P450活性显著高于对照。啶虫脒处理后,马氏管、中肠、脂肪体CarE活性显著增高,马氏管和中肠P450活性显著提高,马氏管GST活性显著提高。乙基多杀菌素处理后,脂肪体和中肠P450活性显著提高,CarE活性在3个组织中均显著提高。高效氯氟氰菊酯处理后,中肠CarE活性显著提高。[结论]6种杀虫剂对东亚飞蝗杀虫活性均较好,其中溴氰虫酰胺生物活性最高。GST、CarE和P450在东亚飞蝗对溴氰虫酰胺、啶虫脒的敏感性中发挥一定作用,CarE和P450可能参与了东亚飞蝗对乙基多杀菌素的解毒代谢,CarE可能是东亚飞蝗对高效氯氟氰菊酯产生耐受力的主要原因。 [Aims] This study aims to explore the toxicity to different insecticides and the effects of different insecticides on the detoxification enzyme activities in Locusta migratoria manilensis(Meyen), in order to provide basis for further study on the resistance management in locust. [Methods] The toxicity of six insecticides to L. migratoria manilensis was determined by leaf immersion method. The effect of LCconcentration of six insecticides on the activity of detoxification enzymes in different tissues of L. migratoria manilensis were determined. [Results] The toxicity of six insecticides to L. migratoria manilensis from high to low was cyantraniliprole, λ-cyhalothrin, acetamiprid, avermectin,imidacloprid and spinetoram. The activity of carboxylesterase(CarE) significantly increased in malpighian tubules,midgut and fat bodies, and glutathione S-transferase(GST) in malpighian tubules and midgut significantly increased after cyantraniliprole treatment. With the treatment of cyantraniliprole, the activity of cytochrome P450 monooxygenase(P450) in malpighian tubules and fat bodies was significantly higher than that of the control. After acetamiprid treatment, the CarE activity in malpighian tubules, midgut and fat bodies, the P450 activity in malpighian tubules and midgut, and the GST activity in malpighian tubules all significantly increased. After spinetoram treatment, the P450 activity in fat bodies and midgut ignificantly increased, and the CarE activity all significantly increased in the three tissues. The CarE activity of midgut significantly increased after treatment with λ-cyhalothrin. [Conclusions] All the six insecticides had good insecticidal activity against L. migratoria manilensis, and cyantraniliprole had the highest toxicity. GST, CarE and P450 may play a certain role in the sensitivity of L. migratoria manilensis to cyantraniliprole and acetamiprid, CarE and P450 may be involved in the detoxification metabolism of spinetoram, and CarE may be the main reason for the tolerance of L. migratoria manilensi
