桃园土壤甲基营养型芽胞杆菌xj-C产脂肽类及挥发性物质的分离与鉴定 被引量:1

摘要 【目的】解析桃园土壤甲基营养型芽胞杆菌xj-C通过次生代谢产生的可能抗菌活性物质种类与成分。【方法】通过PCR扩增和质谱的方法鉴定甲基营养型芽胞杆菌xj-C发酵液的脂肽类物质种类,采用固相微萃取结合气相色谱-质谱联用法对甲基营养型芽胞杆菌xj-C挥发性物质进行分离和鉴定。【结果】甲基营养型芽胞杆菌xj-C发酵液脂肽类物质提取物对桃褐腐病菌有抑菌效果,PCR基因扩增及质谱检测结果发现甲基营养型芽胞杆菌xj-C的脂肽类抑菌活性物质可能是表面活性素C15和泛革素家簇D。甲基营养型芽胞杆菌xj-C释放的挥发性物质对桃褐腐病菌有明显的抑菌作用,甲基营养型芽胞杆菌xj-C有抑菌活性的挥发性物质可能是2,4-二叔丁基苯酚、正十四烷、正十六烷和2-壬酮。【结论】初步分析桃园土壤甲基营养型芽胞杆菌xj-C可能的有效抑菌成分,研究结果为生物菌剂的研发和桃树病害的生物防治提供参考。 【Objective】To analyze the types and components of possible antimicrobial active substances produced by Bacillus methylotrophicu s xj-C through secondary metabolism in peach orchard soil.【Method】The lipopeptides in the fermentation broth of Bacillus methylotrophicu s xj-C were identified by PCR amplification and mass spectrometry.The volatile compounds of Bacillus methylotrophicu s xj-C were isolated and identified by solid-phase microextraction combined with gas chromatography-mass spectrometry.【Result】The lipopeptide extract from the fermentation broth of Bacillus methylotrophicu s xj-C had an inhibitory effect on Monilinia fructicola.PCR gene amplification and mass spectrometry detection results showed that the lipopeptide antibacterial substances of Bacillus methylotrophicu s xj-C might be surface active element C15 and pancretin family D.The volatile compounds released by Bacillus methylotrophicu s xj-C had obvious antibacterial effect on Monilinia fructicola.The volatile compounds with the antibacterial activity of Bacillus methylotrophicu s xj-C may be 2,4-di-tert-butylphenol,tetradecane,hexadecane,and 2-nonanone.【Conclusion】The possible effective antibacterial components of Bacillus methylotrophicu s xj-C in peach orchard soil were preliminarily analyzed,which laid a theoretical foundation for the development of biological agents and the biological control of peach diseases.
