摘要
目的基于PI3K-AKT信号通路探讨免疫球蛋白样受体2(immunoglobulin-like receptor 2,ILR2)是否通过抑制基质金属蛋白酶-2(matrix metalloproteinase-2,MMP-2)影响非小细胞肺癌(non-small cell lung cancer,NSCLC)细胞生物学行为。方法收集2017年9月至2019年3月贵州医科大学附属医院病理科存入的确诊为NSCLC患者的癌组织和癌旁正常肺组织石蜡切片,共48例。采用免疫组织化学染色SP法检测癌组织和癌旁组织中ILR2和MMP-2表达情况。购置NSCLC细胞株,检测各细胞株中ILR2和MMP-2表达水平。转染siRNA敲低ILR2,分析敲低ILR2对H1299细胞增殖、侵袭、转移、凋亡及相关蛋白表达的影响。结果癌组织ILR2和MMP-2阳性表达率显著高于癌旁组织(54.17%比25.00%,58.33%比27.08%,χ^(2)值分别为8.54和9.58,P值均<0.05);H1299细胞株中ILR2和MMP-2表达水平显著高于其他种类NSCLC细胞株,因此将H1299细胞株用于后续研究;沉默组H1299细胞存活率显著低于空白对照组和空载体组[(70.24±13.15)%比(116.24±18.34)%,(70.24±13.15)%比(103.76±20.72)%,t值分别为12.26和11.62,P值均<0.05];沉默组H1299细胞侵袭细胞数目显著少于空白对照组和空载体组[(82.25±9.15)个比(224.18±30.16)个,(82.25±9.15)个比(218.26±28.37)个,t值分别为31.16和34.13,P值均<0.05];沉默组H1299细胞愈合程度显著低于空白对照组和空载体组[(47.26±8.26)%比(60.25±7.25)%,(47.26±8.26)%比(65.24±8.76)%,t值分别为10.25和11.16,P值均<0.05];沉默组H1299细胞凋亡率显著高于空白对照组和空载体组[(29.54±3.17)%比(4.28±1.06)%,(29.54±3.17)%比(3.72±1.01)%,t值分别为7.25和8.16,P值均<0.05];与空白对照组和空载体组比较,沉默组H1299细胞ILR2、MMP-2、磷脂酰肌醇-3激酶(phosphatidylinositol-3 kinase,PI3K)和苏氨酸蛋白激酶(threonine protein kinase,AKT)蛋白表达水平升高,p53蛋白表达水平降低,差异具有统计学意义(t空白对照组=4.03、3.15、3.82、7.26、5.82,t空载体组=2.25、3.26�
Objective To investigate whether the lymphocyte immunoglobulin-like receptor 2(ILR2)has an impact on the biological phenotype of non-small cell lung cancer(NSCLC)cells by the inhibition of matrix metalloproteinase-2(MMP-2)based on the PI3K-AKT signal pathway.Methods Paraffin sections of cancer tissue and adjacent normal lung tissue of 48 patients with NSCLC were collected from the Pathology Department of Affiliated Hospital of Guizhou Medical University from September 2017 to March 2019.The expression of ILR2 and MMP-2 in cancer tissues and adjacent tissues was detected by SP immunohistochemical staining.NSCLC cell lines were purchased to detect the expression levels of ILR2 and MMP-2 in each cell line.Then the effects of silencing ILR2 by transfection of siRNA on the proliferation,invasion,metastasis and apoptosis of H1299 cells and the expression of related proteins were analyzed.Results The positive expression rates of ILR2 and MMP-2 in cancer tissues were significantly higher than those in adjacent tissues(54.17%vs 25.00%,58.33%vs 27.08%,χ^(2) values were 8.54 and 9.58 respectively,both P values<0.05).The expression levels of ILR2 and MMP-2 in H1299 cell line were significantly higher than other types of NSCLC cell lines.Therefore,H1299 cell line was used in the following study.The survival rate of H1299 cells in the silencing group was significantly lower than that in the blank control group and the empty vector group[(70.24±13.15)%vs(116.24±18.34)%,(70.24±13.15)%vs(103.76±20.72)%,t values were12.26 and 11.62 respectively,both P values<0.05].The number of invasive cells of H1299 cells in the silencing group was significantly less than that in the blank control group and the empty vector group[(82.25±9.15)cells vs(224.18±30.16)cells,(82.25±9.15)cells vs(218.26±28.37)cells,t values were 31.16 and 34.13 respectively,both P values<0.05].The healing degree of H1299 cells in the silencing group was significantly lower than that in the blank control group and the empty vector group[(47.26±8.26)%vs(60.25±
