2020⁃2021年南通地区急性病毒性胃肠炎疫情中诺如病毒分子分型分析 被引量:6

Molecular Typing of Noroviruses in An Outbreak of Acute Viral Gastroenteritis in Nantong,China,2020⁃2021

摘要 了解近一年来南通地区急性病毒性胃肠炎疫情的分子流行变异特征。收集2020年6月至2021年5月南通18起疑似病毒性胃肠炎暴发疫情标本共193份,采用实时荧光定量PCR法检测GI/GII组诺如病毒、轮状病毒、札如病毒、星状病毒及肠道腺病毒,对检测结果进行统计学分析。选取其中CT值<30的标本,进行RNA依赖性RNA聚合酶(RNA⁃dependent RNA polymerase,RdRp)区和VP1区序列测定,构建系统进化树。全部193份标本共检出诺如病毒阳性138份,其余标本均为阴性,其中诺如病毒GI组阳性检出率22.5%(31/138),GII组阳性检出率77.5%(107/138);18起疫情基因型别分别为GII.4 Sydney 2012[P16](38.9%,7/18)、GII.3[P12](27.8%,5/18)、GI.3[P13](16.6%,3/18)、GII.4 Sydney 2012[P31](5.6%,1/18)、GII.2[P16](5.6%,1/18)。本研究发现南通市诺如病毒主要流行株为GII.4 Sydney 2012[P16],其他基因型毒株的发现警示需加强监测及病毒测序工作,及时对新型毒株进行检测,进一步收集可用数据,为疾病的防控提供支持。 To understand the molecular epidemic variation characteristics of an outbreak of acute viral gastroenteritis in Nantong in China in the past 12 months.A total of 193 specimens from 18 suspected outbreaks of viral gastroenteritis in Nantong from June 2020 to May 2021 were collected.Samples were analyzed using real⁃time fluorescent quantitative polymerase chain reaction to detect noroviruses of GI/GII genogroups,rotaviruses,sapoviruses,astroviruses,and adenoviruses.CT<30 was selected as a cutoff point.Sequences in the RNA⁃dependent RNA polymerase(RdRp)region and VP1 region were determined,and a system evolution tree was constructed.A total of 138 norovirus⁃positive samples were detected in 193 samples,and the other samples were negative.The positive rate of noroviruses of the GI genogroup was 22.5%(31/138),and that of the GII genogroup was 77.5%(107/138).The genotypes found in 18 outbreaks were GII.4 Sydney 2012[p16](38.9%,7/18),GII.3[p12](27.8%,5/18),GI.3[P13](16.6%,3/18),GII.4 Sydney 2012[P31](5.6%,1/18)and GII.2[p16](5.6%,1/18).The main epidemic strain of noroviruses in Nantong City was GII.4 Sydney 2012(P16).Virus sequencing,timely detection of new strains,and data collection are needed to aid the prevention and control of disease.
