摘要
目的 研究牛樟叶总多糖对大鼠慢性酒精性肝损伤的保护作用与机制。方法 60只雄性大鼠按体质量随机分为对照组、模型组、多烯磷脂酰胆碱胶囊(10.944 mg/kg)组及牛樟叶总多糖高、中、低剂量(50.0、37.5、25.0 mg/kg)组。对照组和模型组给予蒸馏水,其余各组给予不同剂量药物。给药0.5 h后,除对照组外,其余各组均ig体积分数为56%红星二锅头,共给药8周。采用苏木精-伊红(HE)染色观察各组肝组织形态及结构的变化,测定各组大鼠血清白细胞介素-1β(IL-1β)、IL-6、肿瘤坏死因子-α(TNF-α)、丙氨酸基转移酶(ALT)、谷氨酸基转移酶(AST)、三酰甘油(TG)及肝组织匀浆液中IL-1β、IL-6、TNF-α、谷胱甘肽(GSH)水平。免疫组织化学法测定大鼠肝脏沉默信息调节因子1(SIRT1)、磷酸化腺苷酸活化蛋白激酶α(p-AMPKα)表达水平。蛋白质印迹法测定肝组织SIRT1、AMPKα、p-AMPKα、甾醇调节元件结合蛋白1c(SREBP1c)水平。结果 病理切片结果表明,牛樟叶总多糖能改善大鼠肝组织病理变化。与模型组相比,牛樟叶总多糖各剂量组IL-1β、IL-6、TNF-α、TG、ALT、AST水平均明显降低(P<0.05);GSH水平明显升高(P<0.05)。牛樟叶总多糖可提高SIRT1表达及AMPKα的磷酸化水平。结论 牛樟叶总多糖对慢性酒精性肝损伤有一定的保护作用,其机制可能与抗氧化应激、抑制炎症反应、调节SIRT1/AMPK信号通路有关。
Objective To study the protective effect and mechanism of polysaccharide from Cinnamomum camphora leaves on chronic alcoholic liver injury in rats. Methods Sixty male rats were randomly divided into control group, model group, Polyene Phosphatidylcholine Capsules(10.944 mg/kg), polysaccharide from C. camphora leaves high-, medium-and high-dose(50, 37.5 and 25mg/kg) groups according to body weight. Rats in the control group and model group was given distilled water, and rats in the other groups was given corresponding drugs regularly every day. In addition, rats in the treatment groups and model group were given 56%red star erguotou by intragastric administration 0.5 h later to establish chronic alcoholic liver injury model. They were treated for 8weeks. Hematoxylin-eosin(HE) staining was used to observe the changes of liver tissue morphology and structure in each administration group. The levels of IL-1β, IL-6, TNF-α, ALT, AST, and TG in serum and IL-1β, IL-6, TNF-α, and GSH in liver tissue homogenate were determined. The protein levels of SIRT1, AMPKα, P-AMPK α and SREBP1c in liver tissue were determined by Western blotting. The expression levels of SIRT1 and P-AMPK α in rat liver were determined by immunohistochemistry. Results Pathological section results showed that polysaccharide from C. camphora leaves could improve the pathological changes of rat liver tissue. Compared with model group, the levels of IL-1β, IL-6, TNF-α, TG, ALT and AST in all dose groups were significantly decreased(P < 0.05), while the level of GSH was significantly increased(P < 0.05). Total polysaccharide from C. camphora leaves can increase the expression of SIRT1 and phosphorylation of AMPKα. Conclusion Total polysaccharide from C. camphora leaves has a certain protective effect on chronic alcoholic liver injury, and its mechanism may be related to antioxidant stress, antiinflammatory response and regulation of SIRT1-AMPK signaling pathway.
