摘要
目的 探索长托宁对脊髓损伤大鼠细胞焦亡的影响及可能机制。方法 SD大鼠随机分为假手术组(Sham组)、脊髓损伤模型组(Allen’s法建立SCI大鼠模型)和长托宁治疗组(PHC组,腹腔注射长托宁2mg/kg,连续给药14 d)。各组大鼠进行运动功能(BBB)评分;HE、尼氏染色观察脊髓病理学变化;ELISA检测IL-1β、TNF-α、IL-6和IL-18的含量变化;TUNEL检测脊髓细胞凋亡;Western blot方法检测NLRP3、procaspase1、pro-IL1β、IKKβ、IKKα、P-IKKβ、P-IKKα、NF-κB的表达水平。结果 长托宁减轻SCI大鼠脊髓损伤,抑制炎症反应,抑制IL-1β、TNF-α、IL-6和IL-18释放,抑制细胞凋亡,下调炎症小体相关蛋白表达水平,降低NLRP3、pro-caspase1、pro-IL1β表达水平,抑制IKKβ/IKKα/NF-κB信号通路活性。结论 长托宁减轻SCI大鼠炎症反应,抑制NLRP3炎性小体,与调控IKKβ/IKKα/NF-κB通路有关。
Objective To explore the effect and mechanism of penehyclidine hydrochloride on the spinal cord injury and cell pyrolysis in rats. Methods SD rats were randomly divided into sham operation group(Sham group) spinal cord injury model group(SCI, by Allen’s method) and penehyclidine hydrochloride treatment group(PHC, 2 mg/kg intraperitoneally injected for 14 days). HE and Nissl staining were used to observe the pathology of the spinal cord. ELISA was used to detect the content of IL-1β, TNF-α, IL-6 and IL-18. The apoptosis of spinal cord cells were observed by TUNNEL. Western blot was used to detecte NLRP3, pro-caspase1, pro-IL1β,IKKβ, IKKα, P-IKKβ, P-IKKα, NF-κB expressions. Results PHC alleviated the spinal cord injury of SCI rats, inhibited the release of IL-1β, TNF-α, IL-6and IL-18, inhibited apoptosis, and downregulated the levels of NLRP3, pro-caspase1 and pro-IL1β, inhibited the activity of IKKβ/IKKα/NF-κB signaling pathway. Conclusion PHC reduces the inflammatory response and inhibits NLRP3inflammasomes, which is related to the regulation of IKKβ/IKKα/NF-κB pathway in SCI rats.
