基于网络药理学探讨参附汤治疗急性心力衰竭的作用机制 被引量:1

摘要 目的:采用网络药理学技术探讨参附汤治疗急性心力衰竭(Acute Heart Failure,AHF)的作用机制。方法:通过中药系统药理学数据库与分析平台(TCMSP)收集参附汤的化学成分和相关靶点;通过GeneCards、OMIM数据库获取AHF相关疾病靶点;利用String数据库和Cytoscape 3.7.2软件筛选并构建交集靶点相互作用网络;利用R语言对交集靶点进行基因本体论(GO)富集分析和京都基因与基因组百科全书(KEGG)通路富集分析。结果:通过筛选得到参附汤活性成分75个,核心成分为山奈酚、β-谷甾醇、原阿片碱、氟替诺A等;治疗AHF的潜在作用靶点有52个,核心作用靶点包括肿瘤坏死因子(Tumor Necrosis Factor,TNF)、丝氨酸/苏氨酸蛋白激酶1(AKT Serine/Threonine Kinase 1,AKT1)、白细胞介素(Interleukin,IL)-1β、过氧化物酶体增殖剂激活受体γ(Peroxisome Proliferator-activated Receptor Gamma,PPARG)、前列腺素G/H合成酶2(Prostaglandin G/H Synthase 2,PTGS2)。通过富集分析,共获取314条GO富集功能,222条KEGG富集功能,主要与脂质与动脉粥样硬化、糖尿病并发症中晚期糖基化终末产物(Advanced Glycation End Products,AGE)-晚期糖基化终末产物受体(Advanced Glycation End Product Receptor,RAGE)信号通路、流体剪切应力与动脉粥样硬化、TNF信号通路、IL-17信号通路有关。结论:参附汤治疗AHF具有多成分、多靶点和多通路参与的特点,利用网络药理学技术可有效挖掘与分析中药治疗AHF的作用机制。 Objective:To explore action mechanism of the Shenfu decoction(参附汤)on acute heart failure by network pharmacology.Methods:The chemical constituents and related targets of the Shenfu decoction were collected by TCMSP.The targets of AHF-related diseases were obtained through GeneCards and OMIM databases.String database and Cytoscape 3.7.2 software were used to screen out and construct interaction network of intersection targets.The GO enrichment analysis and KEGG pathway enrichment analysis of intersection targets were performed by R language tools.Results:Through screening,75 active components were obtained from the Shenfu decoction,and the core components were kaempferol,β-sitosterol,proopioid,flutinol A,etc..There were 52 potential therapeutic targets for AHF,and the core targets included TNF,AKT1,IL-1β,PPARG and PTGS2.Through enrichment analysis,314 GO enrichment functions and 222 KEGG enrichment functions were obtained,which were mainly related to lipid and atherosclerosis,AGE-RAGE signaling pathway in diabetic complications,fluid shear stress and atherosclerosis,TNF signaling pathway and IL-17 signaling pathway.Conclusion:The Shenfu decoction has the characteristics of multi-component,multi-target and multi-pathway involvement in the treatment of AHF.Network pharmacology technology can be used to explore effectively and analyze the action mechanism of TCM on AHF.
