板蓝根内生产纤维素酶细菌的分离鉴定 被引量:3

摘要 从板蓝根等中药材中分离内生细菌,考察其生物学特性和产纤维素酶性质。依据筛选菌发酵中草药能力及产酶特性,从众多菌株中筛选出产纤维素能力强的菌株,明确其形态及生化特性、适宜生长温度、适宜生长酸碱度、生长曲线、产芽孢能力、药物敏感性和产纤维素能力及适宜条件。结果表明:分离菌为革兰氏阳性芽孢杆菌,经序列测定和同源性分析确定为贝莱斯芽孢杆菌,菌株的最适生长温度为35~40℃,最适生长pH为5.0~8.0,菌株的对数生长期为4~12 h、生长稳定期为12~30 h,72 h时菌株的芽孢形成率为82.1%,菌株的药物敏感性好,培养至36 h时菌株的的纤维素酶活力达31.97 U/mL,温度低于60℃、pH为7.0时菌株的纤维素酶活力比较稳定。分离的贝莱斯芽孢杆菌具有良好的产纤维素酶能力,适宜作为中草药发酵菌种。 To isolate endophytic bacteria from Radix Isatidis and other traditional Chinese medicine,and to investigate their biological characteristics and cellulase producing properties.According to the ability of screening bacteria to ferment traditional Chinese medicine and enzyme production,the strains with strong cellulose production ability were screened from many strains,and their morphological and biochemical characteristics,suitable growth temperature,suitable growth pH,growth curve,spore production ability,drug sensitivity,cellulose production ability and suitable conditions were determined.The results showed that the isolated strain was Gram-positive bacillus.It was identified as Bacillus velezensis by sequence determination and homology analysis.The optimum growth temperature was 35~40℃,the optimum growth pH was 5.0~8.0,the logarithmic growth period of the strain was 4~12 h,the growth stable period was 12~30 h,and the spore formation rate of the strain was 82.1%at 72 h.The cellulase activity of the strain reached 31.97 U/mL when cultured for 36 h.When the temperature was lower than 60℃and p H 7.0,the cellulase activity of the strain was relatively stable.The isolated Bacillus velezensis has good cellulase production ability and was suitable for traditional Chinese medicine fermentation.
