摘要
为研究高粱转录因子BZR1基因家族的表达特征,以高粱BTx623为材料,通过生物信息学及实时荧光定量PCR(quantitative real-time PCR,qPCR)技术对高粱BZR1基因家族进行鉴定和激素应答分析。结果表明,在高粱基因组中共鉴定到9个BZR1基因家族成员,根据染色体位置命名为SbBZR1-1~SbBZR1-9,其氨基酸序列长度在191~716 aa之间,理论分子量大小在20.99~80.41 kD之间,理论等电点在4.75~10.62之间;系统进化树分析显示,高粱BZR1基因家族与单子叶水稻和玉米的同源性为67%~100%;该家族各个成员之间外显子和内含子的数量差异较大。SbBZR1-3、SbBZR1-4和SbBZR1-9基因在高粱不同器官中的表达具有组织特异性;经200µmol/L脱落酸处理后,SbBZR1-3基因的相对表达量逐渐升高,在9 h时达到最大,而高粱SbBZR1-4和SbBZR1-9基因的表达受到显著抑制;SbBZR1-3和SbBZR1-4基因在1µmol/L油菜素内酯诱导下呈降低-升高-降低的表达趋势,但SbBZR1-9基因的表达受到显著抑制;在1 mmol/Lγ-氨基丁酸和100µmol/L褪黑素处理下,SbBZR1-3、SbBZR1-4及SbBZR1-9基因均显著上调表达;经10µmol/L吲哚乙酸处理后,SbBZR1-3基因呈先升高后降低的表达趋势,SbBZR1-9基因的表达受到显著抑制,SbBZR1-4基因呈升高-降低-升高-降低的表达趋势。表明高粱BZR1基因家族在激素应答过程中具有重要作用。
To characterize the expression of transcription factor gene family BZR1 in sorghum,the BZR1 transcription factor family was identified from BTx623 and bioinformatics and quantitative realtime PCR(qPCR)analyses were carried out on their expression hormone responses.The results showed that in the sorghum genome,a total of nine BZR1 family genes were identified,named SbBZR1-1 to SbBZR1-9 according to their chromosome positions.Their amino acid sequence lengths were between 191 and 716,theoretical molecular weights were between 20.99 kD and 80.41 kD,and theoretical isoelectric points were between 4.75 and 10.62.Phylogenetic tree analysis showed that BZR1 transcription factor family showed a homology of 67%to 100%to monocotyledons rice and maize.The analysis of gene structure showed that the number of exons and introns varied greatly among members of the family.The expression of SbBZR1-3,SbBZR1-4 and SbBZR1-9 genes showed tissue specificity in different organs of sorghum.After treatment with 200µmol/L hormone abscisic acid,the relative expression level of SbBZR1-3 was up-regulated gradually and reached the peak at 9 h,while the expressions of SbBZR1-4 and SbBZR1-9 were significantly inhibited.Upon induction of 1µmol/L brassinolide,SbBZR1-3 and SbBZR1-4 genes were down-regulated first,up-regulated late and down-regulated again in 24 h,but the expression of SbBZR1-9 was significantly inhibited.Under the treatment of 1 mmol/Lγ-aminobutyric acid and 100µmol/L melatonin,the expression levels of SbBZR1-3,SbBZR1-4 and SbBZR1-9 genes were significantly up-regulated.After treatment with 10µmol/L indoleacetic acid,the expressions of SbBZR1-3 increased first and then decreased,SbBZR1-9 gene was significantly suppressed,the SbBZR1-4 gene showed an upward-downward-upward-downward trend.These results indicated that sorghum BZR1 family genes play an important role in the hormone responses.
