摘要
肝母细胞瘤(hepatoblastoma,HB)是儿童最常见的肝脏恶性肿瘤,近30年来HB的发病率迅速攀升。目前手术切除和以顺铂为基础的化疗是HB的主要治疗手段。研究表明,Wnt/β-catenin信号通路在肝细胞发生以及肝脏发育、再生和损伤修复等过程中发挥重要作用。同时,Wnt/β-catenin信号通路异常作为HB的主要标志,影响肿瘤的发生发展,且该通路中的一些分子具有作为靶点的潜力。本文对肝母细胞瘤Wnt/β-catenin信号通路的研究进展进行整合,旨在阐明Wnt/β-catenin信号通路的机制及其与HB发生发展的关系,总结Wnt/β-catenin信号通路在HB诊断、治疗、预后等方面的作用,为HB的临床治疗提供参考依据。
Hepatoblastoma(HB)is the most common liver malignancy in children and its incidence has been rising over the last three decades.Surgery excision and cisplatinum-based chemotherapy are currently major treatments for HB.Numerous researches have demonstrated that Wnt signaling pathway plays important roles in hepatocyte generation,liver development,regeneration and injury repair.Meanwhile,abnormal Wnt/β-catenin signaling pathway,as a major marker of HB,affects the occurrence and development of tumors.And some molecules in the pathway have potential targets.By integrating domestic and foreign researches of HB and Wnt/β-catenin signaling pathway,this review elucidated the mechanism of Wnt/β-catenin signaling pathway,examined its relationship with the occurrence and development of HB,summarized the roles of Wnt/β-catenin signaling pathway in the diagnosis,treatment and prognosis of HB and provided novel rationales for clinical treatment of HB.
