Determination of Biotin in Infant Formula Milk Powder by Eppendorf Tube Method

摘要 在GB 5009.259—2016《食品安全国家标准食品中生物素的测定》的基础上,通过缩小培养体系,优化工作菌株的制备和保存方法,调整培养时间等各项措施建立了微生物法检测样品中生物素含量的新方法,并分析了标准曲线线性、精密度、准确度以及加标回收率,验证了方法的实用性,建立了Eppendorf(EP)管法快速检测婴幼儿配方乳粉中生物素质量分数的检测方法。该方法的线性关系良好(r>0.99),精密度(相对标准偏差)为3.27%~4.20%,测定质控样品的相对偏差为–8.8%,加标回收率为90.0%~98.5%。工作菌液和标准工作液在一定的保存条件下1个月内差异不显著(P=0.171),稳定性较好。EP管法操作简便,有效地降低了成本、提高了检测准确性,适合在常规实验室推广和使用。 On the basis of GB5009.259—2016 National food safety standard-Determination of biotin in food,by reducing the culture system,optimizing the method of preparation and preservation of working strains,adjusting the culture time,establish a new method for detecting biotin content in samples by microbial method,and analyze the linearity,precision,accuracy and spiked recovery of standard curve to verify the practicability of the method,establish an Eppendorf(EP)tube method for the rapid determination of biotin content in infant formula milk powder.The linearity of the method was good(r~2>0.99),the relative standard deviations of precision were 3.27%~4.20%,the relative deviation of quality control samples was–8.8%,and the recoveries were 90.0%~98.5%.The difference between working bacteria solution and standard working solution was not significant within 1 month under certain storage conditions(P=0.171),and the stability was good.EP tube method is easy to operate,effectively reduces the cost and improves the detection accuracy.It is suitable to be popularized and used in routine laboratories.
