四妙丸对大鼠痛风性关节炎发展的影响及调控巨噬细胞极化研究 被引量:12

Study on the effect of Simiao Pills on inflammation development and regulation of macrophage polarization in rats with gouty arthritis

摘要 目的:研究四妙丸对尿酸钠晶体(MSU)诱导的大鼠痛风性关节炎发展的影响,并从调控巨噬细胞极化途径探讨其机制。方法:60只大鼠随机分为6组:对照组,模型组,秋水仙碱组及四妙丸高、中、低剂量组,通过踝关节腔内注射MSU制备大鼠痛风性关节炎模型。检测大鼠足肿胀和步态评分,HE染色检测关节组织学评分,ELISA检测血清中白细胞介素(IL)-1β和IL-10表达,Western Blot检测关节组织中诱导型一氧化氮合酶(iNOS)和精氨酸酶1(Arg-1)蛋白表达。结果:与模型组比较,四妙丸中、高剂量组均显著改善了大鼠足肿胀、步态评分和组织学评分(P<0.05,P<0.01);同时亦显著降低了大鼠IL-1β表达(P<0.05),并提高了IL-10表达(P<0.05,P<0.01);四妙丸高剂量组显著降低了iNOS蛋白表达(P<0.01),并提高了Arg-1蛋白表达(P<0.01)。结论:四妙丸可显著抑制大鼠痛风性关节炎发展,其机制与诱导巨噬细胞从M1向M2极化密切相关。 Objective:To investigate the effect of Simiao Pills on monosodium urate-induced arthritis(MIA)and macrophage polarization.Methods:Sixty rats were randomly divided into six groups,which were control,medium,COL and high,medium and low doses of Simiao Pills groups.MIA was prepared by intra-articular injection of MSU crystals in the ankle joint.Paw volume and gait score were measured.Histological score of ankle joints was calculated by HE staining.Levels of IL-1βand IL-10 in serum were detected by ELISA.Expressions of iNOS and Arg-1 were tested by Western Blot.Results:Compared with medium group,high and medium doses of Simiao Pills groups significantly alleviated paw volume,gait score and histological score in MIA rats(P<0.05,P<0.01),meanwhile,high and medium doses of Simiao Pills groups also markedly reduced the levels of IL-1β(P<0.05),and increased the levels of IL-10 in MIA rats(P<0.05,P<0.01),and the high doses of Simiao Pills group markedly reduced the protein expression of iNOS(P<0.01),increased the protein expression of Arg-1 in MIA rats(P<0.01).Conclusion:Simiao Pills might attenuate the MIA development in rats,and the mechanisms are related to induce macrophages from M1 to M2 polarization.
